【摘要】：建立了定量檢測(cè)氟苯尼考的膠體金免疫層析方法。對(duì)膠體金標(biāo)記抗體時(shí)溶液p H和抗體濃度、金標(biāo)抗體用量、檢測(cè)線上抗原濃度以及檢測(cè)時(shí)間進(jìn)行了優(yōu)化。采用膠體金試紙條讀取儀測(cè)定試紙條檢測(cè)線和質(zhì)控線的信號(hào)強(qiáng)度,以標(biāo)準(zhǔn)品的濃度為橫坐標(biāo),陽(yáng)性樣本和陰性樣本的檢測(cè)線/質(zhì)控線的信號(hào)比值(B_x/B_0)為縱坐標(biāo)建立標(biāo)準(zhǔn)曲線。結(jié)果表明,膠體金免疫層析試紙定量檢測(cè)氟苯尼考的線性范圍為0.1~1.5 ng/m L,檢出限為0.08 ng/m L,檢測(cè)時(shí)間為15 min。本方法具有簡(jiǎn)便、快速和可定量等特點(diǎn),適于大批量樣品的現(xiàn)場(chǎng)篩查。
[Abstract]:A colloidal gold immunochromatographic method for quantitative determination of florfenicol was established. The concentration of pH and antibody, the amount of gold labeled antibody, the concentration of antigen on the detection line and the detection time were optimized. The signal strength of test strip and quality control line was measured by colloidal gold strip reader. The standard curve was established by using the concentration of the standard sample as the horizontal coordinate, and the signal ratio of the positive sample and the negative sample (Bx/ B0) as the vertical coordinate. The results showed that the linear range of quantitative determination of florfenicol on colloidal gold immunochromatographic test paper was 0.1 ng/m / L, the detection limit was 0.08 ng/m / L, and the detection time was 15 minutes. The method is simple, rapid and quantifiable, and is suitable for field screening of mass samples.
