溫腎益精散對腎陽虛模型大鼠生長性能及免疫功能影響的研究

【摘要】:生長遲緩是家畜臨床最常見病證之一,中獸醫(yī)學(xué)認(rèn)為生長遲緩與腎陽虛關(guān)系密切。本試驗采用氫化可的松大鼠后肢肌內(nèi)注射構(gòu)建大鼠腎陽虛模型以研究溫腎益精散對其生長性能及免疫功能的影響。試驗將SD大鼠隨機(jī)分為空白對照組、模型組、金匱腎氣湯組、溫腎益精散高、中和低劑量組。記錄各組大鼠體重和采食量變化,計算大鼠飼料轉(zhuǎn)化率,并進(jìn)行血液相關(guān)指標(biāo)測定、免疫器官組織形態(tài)學(xué)觀察,以及肝臟RNA相關(guān)基因表達(dá)的檢測。結(jié)果顯示:1.腎陽虛造?娠@著降低大鼠體重增加速度(P0.05);顯著降低大鼠體溫(P0.05);顯著降低大鼠免疫器官臟器指數(shù)(P0.05);顯著降低大鼠血清中T3、T4、E2、T、ACTH、CORT 和GH含量(P0.05)。2.溫腎益精散中、低劑量組和金匱腎氣湯組可顯著提高模型大鼠體重增加速度(P0.05);顯著提高模型大鼠平均日增重(P0.05);各給藥組均可提高模型大鼠飼料轉(zhuǎn)化率。3.溫腎益精散中、低劑量組和金匱腎氣湯組可顯著降低模型大鼠血清中SST含量(P0.05);顯著提高模型大鼠血清中IGF-1含量(P0.05);溫腎益精散高劑量組可顯著提高模型大鼠血清中IGF-1含量(P0.05);溫腎益精散中劑量組和金匱腎氣湯組可顯著提高模型大鼠肝臟中JAK2和IGF-1mRNA表達(dá)量(P0.05)。4.溫腎益精散中、低劑量組和金匱腎氣湯組可顯著提高模型大鼠胸腺和脾臟臟器指數(shù)(P0.05);溫腎益精散高劑量組可顯著提高模型大鼠胸腺臟器指數(shù)(P0.05);溫腎益精散中劑量組和金匱腎氣湯組可顯著提高模型大鼠血清中IgA、IgG、IgM和IL-2、IL-4含量(P0.05);溫腎益精散低劑量組可顯著提高模型大鼠血清中IL-2和IL-4含量(P0.05);各給藥組均可改善模型大鼠胸腺和脾臟組織形態(tài)變化。結(jié)論:1.溫腎益精散可改善腎陽虛模型大鼠生長性能,提高腎陽虛模型大鼠生長速度。2.溫腎益精散可提高腎陽虛模型大鼠免疫功能。3.溫腎益精散藥理效果以中劑量組(1g/mL)為佳。
[Abstract]:Growth retardation is one of the most common syndromes in animal clinic. Chinese veterinary medicine thinks that growth retardation is closely related to deficiency of kidney yang. A rat model of kidney yang deficiency was established by intramuscular injection of hydrocortisone into hindlimb to study the effect of Wenshen Yijing Powder on its growth performance and immune function. Sprague-Dawley rats were randomly divided into blank control group, model group, Jinkui Shenqi decoction group, Wenshen Yijing San high, medium and low dose groups. The changes of body weight and feed intake were recorded, feed conversion rate was calculated, blood related indexes, morphological observation of immune organs and expression of RNA related genes in liver were measured. The result is 1: 1. Kidney Yang deficiency model can significantly reduce the rate of weight gain in rats (P0.05); significantly decreased the body temperature (P0.05); significantly decreased the organ index of immune organs (P0.05); significantly decreased the serum T3T3OT4E2ACTHCORT and GH content (P0.05). 2. Wenshen Yijing Powder, low dose group and Jingui Shenqi decoction group can significantly increase the weight gain rate of model rats (P0.05); significantly increased the average daily gain of model rats (P0.05); each drug group can increase the feed conversion rate of model rats. 3. Wenshen Yijing Powder, Low dose group and Jinkui Shenqi decoction group can significantly reduce the SST content in the model rat serum (P0.05), significantly increase the IGF-1 content in the model rat serum (P0.05), WYJS high dose group can significantly increase the IGF-1 content in the model rat serum (P0.05); Wenshen Yi-qi decoction group can significantly increase the IGF-1 content in the serum of the model rats (P0.05); Jisan medium dose group and Jinkui Shenqi decoction group could significantly increase the expression of JAK2 and IGF-1 mRNA in the liver of model rats (P0.05). Wenshen Yijing Powder, Low dose group and Jinkui Shenqi decoction group could significantly increase the thymus and spleen organ index (P0.05), WYJS high dose group could significantly increase the thymus organ index (P0.05), WYJS medium dose group and Jinshen Qi decoction group could significantly increase the thymus organ index (P0.05), and Wenshen Yijing Powder moderate dose group and Jinshenqi decoction group could significantly increase the thymus organ index (P0.05). The serum levels of IgAGN IgM and IL-2 IL-4 in model rats were significantly increased (P0.05), IL-2 and IL-4 in serum of model rats were significantly increased in low dose group (P0.05), and the morphologic changes of thymus and spleen of model rats were improved in each administration group. Conclusion 1. Wenshen Yijing Powder could improve the growth performance and increase the growth rate of kidney yang deficiency rats. Wenshen Yijing Powder can improve the immune function of rats with deficiency of kidney yang. The pharmacological effect of Wenshen Yijing Powder was better in the middle dose group (1 g / mL).
