一次性腹腔注射Diquat誘導(dǎo)構(gòu)建肉仔雞慢性氧化應(yīng)激模型

【摘要】:通過(guò)對(duì)肉仔雞一次性腹腔注射Diquat,研究試驗(yàn)期間肉仔雞的生長(zhǎng)性能、部分血液生化指標(biāo)和抗氧化指標(biāo)的變化,建立肉仔雞慢性氧化應(yīng)激模型。選取300只20日齡的健康科寶肉仔雞,隨機(jī)分為2組,每組15個(gè)重復(fù),每重復(fù)10只雞,公母各半。試驗(yàn)期共21d。試驗(yàn)第1天,處理組按照體質(zhì)量以8mg/kg一次性腹腔注射Diquat,對(duì)照組注射相應(yīng)劑量的無(wú)菌生理鹽水。所有雞自由采食和飲水,在試驗(yàn)第1、12、21天,空腹12h,然后前腔靜脈采血分離血清,測(cè)定血清中的生化指標(biāo)和抗氧化指標(biāo)。結(jié)果顯示,Diquat處理組的平均日增量和平均日采食量顯著低于對(duì)照組,料體質(zhì)量比有增高的趨勢(shì);血清中ALT、AST、GSH-Px、SOD和COR活性顯著高于對(duì)照組,MDA濃度顯著低于對(duì)照組。表明,按照8mg/kg體質(zhì)量一次性腹腔注射Diquat溶液可以誘導(dǎo)肉仔雞產(chǎn)生氧化應(yīng)激,并且應(yīng)激效應(yīng)可持續(xù)21d,血清中ALT、AST、GSH-Px、COR、SOD和MDA的含量變化可以作為反映氧化應(yīng)激的敏感指標(biāo)。
[Abstract]:The chronic oxidative stress model of broilers was established by intraperitoneal injection of Diquat in a single dose to study the changes of growth performance, some biochemical indexes and antioxidant indexes of broilers during the experiment. Three hundred 20-day-old healthy Kebao broilers were randomly divided into two groups, 15 replicates in each group, 10 in each replica, half in male and half in female. The trial period was 21 days. On the first day of the experiment, Diquat was injected intraperitoneally with 8mg/kg according to body mass in the treatment group, and the control group was injected with aseptic saline at the corresponding dose. All chickens were fed and drank freely. The serum samples were collected from the anterior vena cava for 12 hours on the 21st day and 12 hours before the first vena cava. The biochemical and antioxidant indexes in the serum were determined. The results showed that the average daily increment and average daily feed intake of Diquat treatment group were significantly lower than those of the control group, and the ratio of feed to body weight was increasing, and the activities of SOD and cor in serum ALT ASTX GSH-PxN were significantly higher than those in the control group, which were significantly lower than those in the control group. The results showed that the oxidative stress of broilers could be induced by single intraperitoneal injection of Diquat solution according to the body weight of 8mg/kg, and the stress effect could be sustained for 21 days. The changes of serum alt ASTX GSH-PxCorn SOD and MDA contents could be regarded as sensitive indexes to reflect oxidative stress.
