【摘要】：為確定桂林某竹鼠養(yǎng)殖場患病竹鼠的病原,從發(fā)病竹鼠肝組織分離到一株細菌。根據(jù)形態(tài)學特征及16S rRNA基因序列分析,確定分離菌株為弗氏檸檬酸桿菌。進化分析顯示,該菌與來自棉花紅蜘蛛的弗氏檸檬酸桿菌遺傳關(guān)系最近,但處于獨立的分支上。毒力試驗結(jié)果表明,該菌對成年小白鼠的LD_(50)為2.8×10~8 CFU,對10~20日齡的竹鼠LD_(50)為6.3×10~8 CFU。藥敏試驗結(jié)果顯示,分離菌株對頭孢曲松、慶大霉素、阿米卡星、氧氟沙星、左氧氟沙星藥物高度敏感,對青霉素、頭孢唑啉、氟苯尼考、林可霉素、強力霉素等有耐藥性。病理組織學觀察顯示,發(fā)病竹鼠肺泡間隔增寬伴隨炎性細胞浸潤;肝組織血管擴張、淤血;脾被膜外大量壞死細胞;腎組織腎小管上皮細胞腫脹、顆粒變性。綜上研究結(jié)果表明,弗氏檸檬酸桿菌是引起桂林竹鼠發(fā)病和致死的病原菌,分離菌株對竹鼠有較強的致病性。
[Abstract]:A bacterial strain was isolated from the liver of a bamboo mouse in a bamboo farm in Guilin. According to morphological characteristics and sequence analysis of 16s rRNA gene, the isolated strain was identified as Citrobacter flexneri. Evolutionary analysis showed that the strain had the closest genetic relationship with Citrobacter flexneri from cotton red spider, but it was an independent branch. The toxicity test showed that the LD50 of the strain was 2.8 脳 10 ~ (8) CFU for adult mice and 6.3 脳 10 ~ (8) CFU for 10 ~ (20) days old bamboo mice. The results showed that the isolates were highly sensitive to ceftriaxone, gentamycin, amikacin, ofloxacin, levofloxacin, and resistant to penicillin, cefazolin, florfenicol, lincomycin and doxycycline. Histopathological observation showed that the alveolar septum widening accompanied by inflammatory cell infiltration; hepatic vasodilation, congestion; a large number of necrotic cells outside the spleen; renal tubular epithelial cells swelling, granular degeneration. The results showed that Citrate flexneri was the pathogen that caused the occurrence and death of bamboo rats in Guilin, and the isolated strains had strong pathogenicity to bamboo rats.
