【摘要】：HH4是新近在荷斯坦牛中發(fā)現(xiàn)的一種遺傳缺陷,其分子機(jī)理為牛1號(hào)染色體上的GART基因編碼區(qū)內(nèi)的1個(gè)A/C錯(cuò)義突變。該遺傳缺陷呈隱性遺傳、突變等位基因純合時(shí),導(dǎo)致胚胎早期死亡。本研究針對(duì)HH4建立了基于PCR-RFLP技術(shù)的分子檢測(cè)方法。結(jié)果表明:通過(guò)對(duì)466頭國(guó)內(nèi)荷斯坦種公牛進(jìn)行基因篩查,發(fā)現(xiàn)6頭攜帶者,均可以追溯到共同的祖先公牛Besne Buck。建議我國(guó)今后進(jìn)口荷斯坦種牛時(shí),對(duì)個(gè)體進(jìn)行系譜分析和遺傳缺陷基因分子檢測(cè),避免引入有害基因;同時(shí),對(duì)國(guó)內(nèi)現(xiàn)有荷斯坦公牛群體進(jìn)行HH4篩查,明確標(biāo)注每頭公牛的攜帶狀態(tài),以便在生產(chǎn)實(shí)踐中合理選種選配。
[Abstract]:HH4 is a newly discovered genetic defect in Holstein cattle. Its molecular mechanism is an A / C missense mutation in the coding region of the Gart gene on chromosome 1. The genetic defect is recessive and causes early embryo death when the mutant allele is homozygous. In this study, a molecular detection method based on PCR-RFLP was established for HH 4. The results showed that 6 carriers were found by genetic screening of 466 domestic Holstein bulls, all of which could be traced back to Besne Buckle, a common ancestor bull. In order to avoid the introduction of harmful genes, we should carry out pedigree analysis and genetic defect gene molecular detection in order to avoid the introduction of harmful genes. At the same time, we should carry out HH4 screening for the existing Holstein bull population in China. The carrying status of each bull is clearly marked in order to select species and match reasonably in production practice.
