寧海黃雞和廣西黃雞KLF7基因多態(tài)性及其與屠體性狀的相關(guān)分析

【摘要】:Kruppel樣因子7(Kruppel-like factor,KLF7)是參與脂肪代謝的一個(gè)因子,其對(duì)雞(Gallus gallus)生長(zhǎng)和屠體性狀的相關(guān)研究較少。本研究采用600K Affymetrix Axiom Chicken SNP芯片技術(shù),研究雞KLF7基因單核苷酸多態(tài)性(SNPs)及其與屠體性狀的相關(guān)性。本實(shí)驗(yàn)經(jīng)對(duì)寧海黃雞和廣西黃雞各59個(gè)個(gè)體進(jìn)行基因分型,在KLF7基因上共發(fā)現(xiàn)38個(gè)SNPs位點(diǎn),其中4個(gè)SNPs位點(diǎn)位于外顯子上。除寧海黃雞T16936C、A18000C、C33569T、T34758C和T40474G位點(diǎn)以及廣西黃雞A18715C和T45125A位點(diǎn)外,其余位點(diǎn)均處于Hardy-Weinberg平衡狀態(tài),多態(tài)信息含量顯示,寧海黃雞和廣西黃雞KLF7基因大多數(shù)位點(diǎn)屬中度多態(tài)。各SNPs位點(diǎn)與寧海黃雞和廣西黃雞屠體性狀相關(guān)性分析結(jié)果表明,分別有7個(gè)和6個(gè)SNPs位點(diǎn)對(duì)部分屠體性狀有顯著或極顯著影響(P0.05或P0.01)。其中,A18000C和T45125A兩個(gè)SNPs位點(diǎn)對(duì)寧海黃雞和廣西黃雞屠體性狀均有顯著或極顯著影響(P0.05或P0.01)且寧海黃雞有AA、AB和BB 3種基因型,而廣西黃雞僅有AB和BB兩種基因型。寧海黃雞A18000C位點(diǎn)AB型的胸肌率極顯著高于BB型(P0.01),T45125A位點(diǎn)AB型的半凈膛率和全凈膛率極顯著高于AA型(P0.01),顯著高于BB型(P0.05),AB型的腹脂重和腹脂率顯著高于AA型和BB型(P0.05);廣西黃雞A18000C位點(diǎn)AB型的胸肌率顯著高于BB型(P0.05),T45125A位點(diǎn)AB型的屠宰率、半凈膛率、全凈膛重和全凈膛率顯著高于BB型(P0.05)。研究結(jié)果表明,KLF7基因?qū)﹄u屠體性狀有一定的基因效應(yīng),為雞屠體性狀具有潛力的候選基因,可作為雞屠體性狀的分子標(biāo)記輔助選擇。
[Abstract]:Kruppel-like factor 7 (KLF7) is one of the factors involved in fat metabolism. There are few studies on the relationship between growth and carcass traits in chicken (Gallus gallus). The single nucleotide polymorphism (SNPs) of chicken KLF7 gene and its association with carcass traits were studied by 600K Affymetrix Axiom Chicken SNP chip technique. In this study, 38 SNPs loci were found on the KLF7 gene by genotyping of 59 individuals in Ninghai Yellow Chicken and Guangxi Yellow Chicken respectively, and 4 SNPs loci were found on exon. With the exception of T16936CA18000 C33569THN T34758C and T40474G, and Guangxi Yellow Chicken A18715C and T45125A, all the other loci were in Hardy-Weinberg equilibrium. The polymorphic information content showed that the majority of KLF7 loci in Ninghai chicken and Guangxi yellow chicken were moderately polymorphic. The results of correlation analysis between SNPs loci and carcass traits of Ninghai Yellow Chicken and Guangxi Yellow Chicken showed that 7 SNPs loci and 6 SNPs loci had significant or extremely significant effects on some carcass traits (P0.05 or P0.01). The two SNPs loci of A18000C and T45125A had significant or extremely significant effects on carcass traits of Ninghai Yellow Chicken and Guangxi Yellow Chicken (P0.05 or P0.01), and Ninghai Yellow Chicken had three genotypes of AAAAB and BB, while Guangxi Yellow Chicken had only AB and BB genotypes. The breast muscle rate of A18000C genotype AB in Ninghai yellow chicken was significantly higher than that of BB type (P0.01) and AB type (P0.01), and the percentage of total purge was significantly higher than that of AA type (P0.01), and was significantly higher than that of BB type (P0.05), abdominal fat weight and abdominal fat rate of BB type AB (P0.05), while that of BB type (P0.01) was significantly higher than that of AA type and BB type (P0.05). The breast muscle rate of A18000C genotype AB was significantly higher than that of BB type (P0.05), and the slaughter rate of AB type at T45125A locus was significantly higher than that of BB type (P0.05). The rate of half-bore, full-bore weight and total-purged bore was significantly higher than that of BB-type (P 0. 05). The results showed that KLF7 gene had a certain gene effect on chicken carcass traits, which was a candidate gene with potential for chicken carcass traits and could be used as a molecular marker assisted selection for chicken carcass traits.
