【摘要】：谷胱甘肽過氧化物酶-5 (Glutathione peroxidase type-5)在動物附睪中特異性表達(dá),并對精子的成熟起著至關(guān)重要的作用。本文研究了GPX5在不同年齡的綿羊附睪中的表達(dá)情況。研究結(jié)果如下：1.通過實(shí)時熒光定量PCR,檢測GPX5基因在不同年齡綿羊附睪,睪丸和輸精管的相對表達(dá)量。結(jié)果表明GPX5基因在幼齡(2-3月齡)羔羊附睪、睪丸和輸精管中幾乎不表達(dá)；而在青年(6-12月齡)綿羊附睪頭,GPX5基因的表達(dá)量極顯著高于附睪體和附睪尾(p0.01),在輸精管和睪丸幾乎不表達(dá)；在成年(2-3歲)綿羊附睪頭顯著高于附睪體和附睪尾(p0.05),但顯著低于青年綿羊附睪頭的表達(dá)量。GPX5在不同年齡綿羊的睪丸和輸精管的表達(dá)量沒有顯著差異。2.通過western blot的方法檢測不同年齡綿羊的附睪和睪丸中GPX5蛋白的表達(dá)。結(jié)果顯示在羔羊附睪和睪丸未檢測到GPX5蛋白；在青年綿羊附睪頭高表達(dá),附睪體和附睪尾中低表達(dá),睪丸中不表達(dá)；在成年綿羊附睪頭少量表達(dá),附睪體,附睪尾和睪丸幾乎都不表達(dá)。3.利用免疫組化的方法,對GPX5蛋白在附睪上進(jìn)行定位。經(jīng)過免疫組化分析,GPX5蛋白主要在附睪頭上皮細(xì)胞質(zhì)以及靜纖毛表達(dá),附睪體和附睪尾中的GPX5蛋白主要集中在附睪管腔中,上皮少量表達(dá)。并且附睪尾的GPX5蛋白主要在靜纖毛表達(dá)。GPX5蛋白從附睪上皮分泌到附睪腔中,隨著精子在附睪中的運(yùn)輸與精子結(jié)合�？傊�,GPX5在不同年齡的綿羊附睪中的表達(dá)存在差異。GPX5主要在青年綿羊和,成年綿羊的附睪頭上皮表達(dá),GPX5蛋白分泌到附睪管腔中與精子結(jié)合,保護(hù)精子在附睪運(yùn)輸過程中不受活性氧(ROS)的攻擊,保證精子功能的正常發(fā)揮。
[Abstract]:Glutathione peroxidase -5 (Glutathione peroxidase type-5) is specifically expressed in animal epididymis and plays a vital role in the maturation of sperm. This paper studies the expression of GPX5 in the epididymis of sheep at different ages. The results are as follows: 1. by real-time quantitative fluorescence quantitative PCR, the GPX5 gene was detected at different ages. The relative expression of the epididymis, testis and vas deferens showed that the GPX5 gene was almost not expressed in the epididymis of young (2-3 months old) lambs, testis and vas deferens, while the expression of GPX5 gene in the epididymal head of the young (6-12 month old) sheep was significantly higher than that of the epididymis and epididymal tail (P0.01), and was almost not expressed in the spermatic duct and testis; in adulthood (2-3) The epididymal head of sheep was significantly higher than the epididymal body and epididymal tail (P0.05), but it was significantly lower than the expression of.GPX5 in the epididymis of the young sheep. In a word, the expression of GPX5 in the epididymis of sheep in different ages is different.GPX5 mainly in the young sheep and the epithelia of the epididymis of adult sheep, GPX5 protein secreted into the epididymal cavity with sperm, protecting sperm from the attack of active oxygen (ROS) during the epididymal transport, so as to ensure the normal functioning of the sperm function.
