犬MG53基因的克隆與表達

【摘要】:為進一步研究犬MG53在機體內的分布情況及其生物活性,本研究設計特異性引物進行犬MG53目的基因的克隆,并將其連接至p MAL-C5X表達載體中,利用大腸桿菌表達系統(tǒng)進行表達。結果顯示,克隆得到由1 435個核苷酸構成的犬MG53基因;獲得MG53蛋白與麥芽糖結合蛋白標簽(MBP)融合表達的可溶性重組蛋白,大小約為95 ku。通過結構及生物學功能預測發(fā)現,該蛋白易降解,具有RING、B-Box、Coiledcoil及SPRY-PRY四個功能結構域,并分布有半胱胺(Cysteamine)、2-氨基乙硫醇(2-Amino-1-Ethanethiol)、硫代乙醇胺(Thioethanolamine)、2,3-Deshydrolanthionine、Zn2+及Mg2+等分子結合位點,具有蛋白結合能力。上述結果表明,犬MG53與人、家兔、小鼠、大鼠MG53的結構相似,并可能具有參與泛素化、骨骼肌細胞修復等重要的生物學功能。
[Abstract]:In order to further study the distribution and biological activity of dog MG53, a specific primer was designed to clone the target gene of dog MG53, which was ligated into pMAL-C5X expression vector and expressed by E. coli expression system. The results showed that the canine MG53 gene composed of 1,435 nucleotides was cloned and the soluble recombinant protein expressed by fusion of MG53 protein and maltose binding protein label (MBP) was obtained, the size of which was about 95 ku. It was found that the protein was easy to degrade and had four functional domains RINGB-Boxen Coiledcoil and SPRY-PRY. The protein was characterized by cysteamine 2-aminoethiol thioethanolamine 2 3-Deshydrolanthionine Zn2 and Mg2. These results suggest that the structure of dog MG53 is similar to that of human, rabbit, mouse and rat MG53, and may have important biological functions such as ubiquitization, skeletal muscle cell repair and so on.
