【摘要】：為研究牛血紅蛋白源抗菌肽P3及其類似肽(JH-0、JH-1、JH-2、JH-3)的生物學(xué)功能,本研究利用在線軟件分析了抗菌肽P3及其類似肽的生物信息學(xué)特性,發(fā)現(xiàn)這5個抗菌肽均帶正電荷,等電點(diǎn)均大于8.00,均為疏水蛋白,定位于細(xì)胞外,不含有跨膜區(qū)、信號肽序列和糖基化位點(diǎn),P3和JH-2的二級結(jié)構(gòu)為α-螺旋+β-折疊,JH-3為100%的α-螺旋,JH-0和JH-1為無規(guī)則卷曲結(jié)構(gòu),P3、JH-2和JH-3在第13位氨基酸即蘇氨酸(Thr,T)有一個糖基化位點(diǎn)。根據(jù)分析結(jié)果推測抗菌肽P3及其類似肽可作用于細(xì)胞壁或細(xì)胞膜,從而使菌體死亡。
[Abstract]:In order to study the biological function of bovine hemoglobin derived antimicrobial peptide P3 and its analogous peptide JH-0, the bioinformatics characteristics of antimicrobial peptide P3 and its analogous peptides were analyzed by on-line software. It was found that the five antimicrobial peptides were positively charged. The isoelectric points were more than 8.00, they were hydrophobic proteins, which were located outside the cells and did not contain the transmembrane region. The signal peptide sequence and glycosylation sites P3 and JH-2 have a glycosylation site at the 13th amino acid, namely, threonine thr (THR) with 100% 偽 -helix JH-0 and JH-1 as irregularly curled structure (P _ 3H _ 2 and JH _ 3), and the secondary structures of the signal peptide and glycosylation sites are 偽 -helix 尾 -foldout (JH-3) and JH-1 (100%). The results suggested that the antibacterial peptide P3 and its analogous peptides could act on the cell wall or cell membrane, resulting in the cell death.
