黑絲羽烏骨雞精液DMA顆粒冷凍技術(shù)研究

【摘要】:為研究不同稀釋液與解凍裝置對(duì)黑絲烏骨雞N,N-二甲基甲酰胺(N,N-dimethylformamide,DMA)顆粒凍精解凍后精子質(zhì)量的影響,試驗(yàn)首先采用不同稀釋液對(duì)精液進(jìn)行稀釋并比較其解凍后精子活力與人工輸精的受精率;其次,選取不同解凍管及凍精顆粒數(shù)進(jìn)行恒溫水浴解凍,比較其活力;最后,依據(jù)解凍后的精子活力,篩選恒溫水浴、恒溫漏斗、恒溫板3種解凍裝置各自的最佳解凍溫度,并利用各自最佳解凍溫度解凍后的精液進(jìn)行人工輸精,檢測(cè)受精率。結(jié)果顯示:1不同稀釋液組的精子活力與受精率高低趨勢(shì)一致,為L(zhǎng)RFBL組,且各組之間差異顯著(P0.05)。2在60℃恒溫水浴中,用薄壁大玻璃管解凍的精子活力最好。3不同解凍裝置有各自最佳解凍活力的溫度范圍,恒溫水浴為50~60℃(0.51~0.59)、恒溫漏斗為40~45℃(0.42~0.46)、恒溫板為50~55℃(0.61~0.63),每種裝置最佳溫度段內(nèi)的精子活力差異不顯著(P0.05)。43種解凍裝置最佳解凍狀態(tài)相比:在精子活力上,60℃恒溫水浴與55℃恒溫板分別顯著高于40℃恒溫漏斗(P0.05),但60℃恒溫水浴與55℃恒溫板之間差異不顯著(P0.05);在受精率上,55℃恒溫板最高(26.91%),60℃恒溫水浴次之(23.08%)、40℃恒溫漏斗最低(20.93%),三者之間差異不顯著(P0.05)。因此,黑絲羽烏骨雞精液應(yīng)采用LR稀釋液、DMA冷凍保護(hù)劑及顆粒冷凍技術(shù),在54.9℃恒溫板解凍可獲得較高的受精率。
[Abstract]:In order to study the effects of different diluents and thawing devices on the sperm quality of NN- dimethylformamide (DMA) granules of black silk silky chicks after thawing, At first, different diluents were used to dilute semen and compare the fertilization rate between sperm motility and artificial insemination after thawing. Secondly, different thawing tubes and the number of frozen sperm granules were selected for constant temperature water bath thawing to compare their motility. According to the sperm motility after thawing, the optimum thawing temperature of the three thawing devices, namely, the constant temperature water bath, the constant temperature funnel and the constant temperature plate, were selected, and the semen after thawing at their optimum thawing temperature were used for artificial insemination to detect the fertilization rate.
