大通牦牛心肌低氧適應(yīng)的組織學(xué)特點

【摘要】:選取兩個海拔高度大通牦牛作為研究對象,以樂都地區(qū)牦牛為對照,利用光鏡技術(shù)和計算機圖像分析系統(tǒng)測定心肌肌纖維直徑、表面積密度;利用透射電鏡技術(shù)比較心肌線粒體的平均體積(V)、體積密度(VV)和面數(shù)密度(NA)等結(jié)構(gòu)參數(shù);利用免疫組織化學(xué)技術(shù)對心肌細胞血管內(nèi)皮生長因子(VEGF)和微血管密度(MVD)進行檢測,并對以上數(shù)據(jù)進行相關(guān)性分析。結(jié)果顯示:(1)大通牦牛心肌肌纖維直徑均細于樂都地區(qū)牦牛心肌肌纖維直徑,肌纖維表面積密度均高于樂都地區(qū)牦牛,且三者兩兩比較差異顯著(P0.05);(2)大通牦牛的VEGF和MVD均高于樂都地區(qū)牦牛,差異顯著(P0.05);(3)大通牦牛心肌線粒體的V和VV均大于樂都地區(qū)牦牛,差異顯著(P0.05);海拔3 700m的大通牦牛心肌線粒體面數(shù)密度大于海拔3 200m的樂都地區(qū)牦牛,差異顯著(P0.05)。結(jié)果表明,大通牦牛心肌組織主要表現(xiàn)為心肌纖維直徑細、表面積密度大、心肌組織血管內(nèi)皮生長因子和微血管密度大的遺傳學(xué)特點。而心肌線粒體的低氧適應(yīng)表現(xiàn)為,隨著海拔高度的增加,心肌線粒體平均體積增大,面數(shù)密度和體密度增加的特點。
[Abstract]:The diameter and surface density of cardiac muscle fiber were measured by light microscope and computer image analysis system. The structural parameters of myocardial mitochondria were compared by transmission electron microscopy (TEM), mean volume density (VV) and surface number density (NAA), and vascular endothelial growth factor (VEGF) and microvessel density (MVD) were detected by immunohistochemistry. And carries on the correlation analysis to the above data. The results showed that the diameter of cardiac muscle fiber in Datong yak was smaller than that in Ledu area, and the density of muscle fiber surface area was higher than that in Ledu area. The difference of VEGF and MVD of Datong yak was higher than that of Yak in Ledu area, and the difference was significant (P 0.05). The V and VV of myocardial mitochondria of Datong yak were higher than that of Yak in Ledu area. The difference was significant (P 0.05), and the density of mitochondria surface of Datong yak at 3 700 m above sea level was higher than that of Yak in Ledu area at 3 200 m above sea level, the difference was significant (P 0 05). The results showed that the myocardial tissue of Datong yak was characterized by fine diameter of myocardial fiber, large density of surface area, high vascular endothelial growth factor (VEGF) and microvessel density. However, the hypoxic adaptation of myocardial mitochondria was characterized by the increase of the average volume of myocardial mitochondria, the increase of surface number density and volume density with the increase of altitude.
