初情期啟動(dòng)過(guò)程中MeCP2和GnRH在母羊下丘腦的分布定位

【摘要】:本實(shí)驗(yàn)旨在研究甲基化CpG結(jié)合蛋白(MeCP2)和促性腺激素釋放激素(GnRH)初情期啟動(dòng)過(guò)程中在母羊下丘腦的分布及二者關(guān)系。選取初情期前、臨近初情期和初情期階段雌性小尾寒羊各3只,采集下丘腦,利用免疫熒光雙標(biāo)記技術(shù)研究MeCP2和GnRH在母羊下丘腦視前區(qū)(POA)和弓狀核(ARC)中的分布。結(jié)果表明:MeCP2和GnRH在下丘腦ARC和POA均有分布,且呈現(xiàn)共表達(dá)模式,ARC中共表達(dá)陽(yáng)性細(xì)胞數(shù)在初情期顯著高于初情期前期和臨近初情期(P0.05),POA中共表達(dá)陽(yáng)性細(xì)胞數(shù)初情期顯著低于初情期前期和臨近初情期(P0.05)。結(jié)果提示,MeCP2可能通過(guò)調(diào)控下丘腦GnRH的表達(dá)而參與母羊初情期啟動(dòng)的調(diào)控。
[Abstract]:The aim of this study was to investigate the distribution of methylated CpG-binding protein (MeCP2) and gonadotropin releasing hormone (GnRH) in the hypothalamus of ewe during the initial estrus. The hypothalamus was collected from three female small tail Han sheep before, near and near estrus, and the distribution of MeCP2 and GnRH in preoptic area (POA) and arcuate nucleus (ARC) of hypothalamus were studied by immunofluorescence double labeling technique. The results showed that both of them were distributed in the hypothalamus ARC and POA. The number of co-expression positive cells of ARC in the early estrus was significantly higher than that in the early estrus and near the early estrus, and the number of P0.05 POA co-positive cells in the early estrus was significantly lower than that in the early estrus and near estrus. The results suggest that MeCP2 may be involved in the regulation of the initiation of ewe at the beginning of estrus by regulating the expression of GnRH in the hypothalamus.
