【摘要】:為評價(jià)禽多殺性巴氏桿菌ptfA基因殼聚糖納米DNA疫苗的免疫效果,本實(shí)驗(yàn)以殼聚糖為佐劑制備ptfA基因的殼聚糖納米DNA疫苗,檢測其形態(tài)及抗DNA酶降解的能力,隨后進(jìn)行動物免疫實(shí)驗(yàn),檢測免疫后小鼠血清特異性抗體水平、免疫小鼠脾淋巴細(xì)胞增殖、IFN-γ分泌情況和抵抗強(qiáng)毒菌株攻擊和免疫保護(hù)率。實(shí)驗(yàn)結(jié)果顯示,ptfA基因殼聚糖納米DNA在透射電鏡下呈現(xiàn)較為規(guī)則的圓球型,粒徑約為200 nm,可以有效抵抗DNAaseⅠ的降解。間接ELISA檢測結(jié)果顯示,殼聚糖納米DNA疫苗組和裸DNA組的血清抗體水平均呈現(xiàn)上升趨勢,明顯高于空載體對照組和PBS對照組,且殼聚糖納米DNA疫苗組的抗體水平稍高于裸DNA疫苗組。淋巴細(xì)胞增殖試驗(yàn)和IFN-γ分泌試驗(yàn)結(jié)果也顯示,殼聚糖納米DNA疫苗組和裸DNA疫苗組的SI值與IFN-γ分泌水平極顯著高于兩對照組(p0.01),但兩種DNA疫苗組之間差異并不顯著(p0.05)。強(qiáng)毒菌株攻擊后殼聚糖納米DNA疫苗組的保護(hù)率優(yōu)于裸DNA疫苗組,在一定程度上增強(qiáng)了禽多殺性巴氏桿菌ptfA基因裸DNA疫苗的免疫效果。從而為禽多殺性巴氏桿菌新型DNA疫苗的研究奠定了基礎(chǔ)。
[Abstract]:In order to evaluate the immune effect of ptfA gene chitosan nano DNA vaccine of Pasteurella multocida, chitosan nano DNA vaccine containing ptfA gene was prepared with chitosan as adjuvant, and its morphology and ability to resist the degradation of DNA enzyme were tested. Then, the level of specific antibody in serum, the secretion of IFN- 緯 in spleen lymphocytes of immunized mice, the rate of immunity against the attack of virulent strains and the rate of immune protection were detected. The experimental results showed that the chitosan nanocrystalline DNA with ptfA gene exhibited a regular spherical shape under transmission electron microscope with a diameter of about 200nm. it could effectively resist the degradation of DNAase 鈪,
