【摘要】：為探討12個(gè)長(zhǎng)鏈非編碼RNA(lnc RNA)在發(fā)生炎癥反應(yīng)的仔豬骨骼肌和肝臟組織炎癥反應(yīng)中的差異表達(dá),本研究選取12頭(28±3)日齡、體重(9.0±0.7)kg的杜×長(zhǎng)×大斷奶仔豬,分成2個(gè)處理,每個(gè)處理6個(gè)重復(fù);2個(gè)處理組基礎(chǔ)飼糧相同,試驗(yàn)第21天,LPS組腹膜注射100μg/kg體重(BW)的LPS,對(duì)照組注射等量的生理鹽水。于注射LPS或生理鹽水4 h后屠宰,取骨骼肌和肝臟樣品待測(cè)。結(jié)果表明:LPS刺激導(dǎo)致骨骼肌和肝臟組織中腫瘤壞死因子-α(TNF-α)、環(huán)氧合酶2(COX2)和核苷酸結(jié)合寡聚域受體2(NOD2)的m RNA表達(dá)量顯著提高(P0.05);LPS刺激極顯著降低骨骼肌中l(wèi)nc RNA-36199.1的表達(dá)量(P0.01);LPS刺激顯著提高肝臟中l(wèi)nc RNA-MEG3、lnc RNA-RP11-451G4.2、lnc RNA-27609.1、lnc RNA-32021.1、lnc RNA-46606.2和lnc RNA-36199.1的表達(dá)量(P0.05),但顯著降低lnc RNA-30795.1和lnc RNA-26244.1的表達(dá)量(P0.05)。本試驗(yàn)研究表明,8種lnc RNA在LPS誘導(dǎo)的斷奶仔豬骨骼肌或肝臟的炎癥反應(yīng)中存在差異表達(dá),說(shuō)明其可能參與不同組織的炎癥反應(yīng)。
[Abstract]:In order to investigate the differential expression of 12 long chain noncoding RNA(lnc RNAs in skeletal muscle and liver tissues of piglets with inflammatory response, 12 piglets with 9. 0 鹵0.7)kg body weight and 28 鹵3 days of age were selected and divided into 2 treatments. Two treatment groups received the same basal diet. On the 21st day of the experiment, LPSs of 100 渭 g/kg BW were injected into the peritoneum of LPS-treated group and the control group was injected with the same amount of normal saline. Four hours after injection of LPS or normal saline, skeletal muscle and liver samples were slaughtered. The results showed that the expression of m RNA of TNF- 偽 TNF- 偽, cyclooxygenase-2 (COX2) and nucleotide-binding oligodeoxyribonucleotide receptor (2nd2) in skeletal muscle and liver was significantly increased by the stimulation of TNF- 偽 TNF- 偽, COX-2) and the expression of lnc RNA-36199.1 in skeletal muscle by the stimulation of lipopolysaccharide (LPS). The expression of lnc RNA-MEG3LNC RNA-RP11-451G4.2LNC RNA-27609.1lncRNA-32021.1lnc RNA-46606.2 and lnc RNA-36199.1 in liver was significantly increased by LPS stimulation, but the expression of lnc RNA-30795.1 and lnc RNA-26244.1 was significantly decreased (P0.05). This study showed that eight lnc RNA were differentially expressed in skeletal muscle or liver of weaned piglets induced by LPS, suggesting that they may participate in the inflammatory response of different tissues.
