運(yùn)用高通量蛋白質(zhì)組學(xué)方法鑒定與PRRSV NSP7結(jié)合的宿主細(xì)胞蛋白

【摘要】:豬繁殖與呼吸系統(tǒng)綜合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)雖已被國內(nèi)外科研工作者廣泛研究,但其中一些非結(jié)構(gòu)蛋白例如NSP7的結(jié)構(gòu)和功能依然所知甚少,據(jù)推測此蛋白對病毒的RNA合成和蛋白翻譯過程十分重要。本試驗(yàn)運(yùn)用蛋白質(zhì)組學(xué)技術(shù)偶聯(lián)免疫共沉淀法,并結(jié)合生物信息學(xué)方法分析,鑒定出12種與PRRSV NSP7產(chǎn)生直接或間接相互作用的宿主細(xì)胞蛋白,NSP7與這些蛋白之間的互作關(guān)系能夠?yàn)镹SP7的深入研究提供新的途徑和思路。
[Abstract]:Porcine reproductive and respiratory syndrome virus (PRRSVV) has been widely studied by researchers at home and abroad, but the structure and function of some non-structural proteins such as NSP7 are still unknown. It is speculated that this protein is very important in RNA synthesis and protein translation. In this study, proteomics coupled immunoprecipitation and bioinformatics analysis were used. Twelve kinds of host cell proteins that interact directly or indirectly with PRRSV NSP7 have been identified. The interaction between NSP7 and these proteins can provide new approaches and ideas for the further study of NSP7.
