蛋雞感染NDV后輸卵管組織中CaBP-D28k、OVA基因mRNA表達(dá)及蛋品質(zhì)量變化研究

【摘要】:新城疫是由新城疫病毒(NDV)引起禽類的一種急性、高度傳染性疾病。NDV感染產(chǎn)蛋雞群,除發(fā)生發(fā)病死亡外,還可引起生殖系統(tǒng)病變,導(dǎo)致雞群產(chǎn)蛋量和蛋品質(zhì)下降,產(chǎn)沙殼蛋、軟殼蛋和畸形蛋等。但目前有關(guān)NDV感染蛋雞后導(dǎo)致輸卵管組織病變與蛋品質(zhì)下降的機(jī)制還不清楚。本研究通過滴鼻、點(diǎn)眼途徑對240日齡非免疫健康羅曼白蛋雞感染NDV/Chicken/TC/1/2011株,在健康對照雞和感染后第1、3、5、7、9、11、15天,收集健康對照雞和感染雞所產(chǎn)雞蛋檢測蛋殼質(zhì)量,采用原子分光光度法檢測蛋殼中鈣含量,采用掃描電子顯微鏡觀察雞蛋蛋殼內(nèi)、外表面和橫斷面超微結(jié)構(gòu);在健康對照組和感染后第1、3、5、7、9、11、15天,分別撲殺一定數(shù)量雞,無菌采集輸卵管膨大部、子宮部,利用實(shí)時(shí)熒光定量PCR檢測與蛋殼鈣化相關(guān)的CaBP-D28k基因和與蛋清質(zhì)量相關(guān)的卵清蛋白(OVA)基因mRNA的表達(dá)變化,結(jié)果如下:1.蛋雞感染NDV后1、3、5、7、9、11、15天,感染雞所產(chǎn)雞蛋蛋殼中鈣的平均相對含量(質(zhì)量比)分別40.60%、38.41%、34.15%、37.42%、38.58%、39.03%、39.50%,健康對照雞為41.88%;感染雞所產(chǎn)雞蛋蛋殼橫斷面平均厚度分別305um、290um、279.5um、313um、320.50um、321um、323um,健康對照雞為368.5um。感染NDV后1~5天,雞蛋乳突層變薄,柵欄層和垂直晶體層的平整性和致密性均不同程度地下降;蛋殼外表面的角質(zhì)層逐漸變粗糙,氣孔增大,皸裂明顯;蛋殼內(nèi)表面纖維間隙逐漸增大,結(jié)構(gòu)稀疏、斷裂明顯、其上的結(jié)點(diǎn)明顯變少。感染NDV后7~15天,蛋雞所產(chǎn)雞蛋蛋殼質(zhì)量緩慢改善。2.蛋雞感染NDV后1~5天,感染雞產(chǎn)軟殼蛋、薄殼蛋,輸卵管子宮部CaBPD28k基因mRNA表達(dá)量持續(xù)下降,感染第5天時(shí)其表達(dá)量僅為健康對照雞的0.16倍;感染后7~15天,發(fā)病雞產(chǎn)蛋蛋殼質(zhì)量逐漸改善,子宮部CaBP-D28k基因mRNA表達(dá)量逐漸回升。感染后第15天,子宮部CaBP-D28k基因mRNA表達(dá)量達(dá)到健康對照雞的0.45倍。3.蛋雞感染NDV后1、3、5、7、9、11、15天,感染雞所產(chǎn)雞蛋蛋清中蛋白相對含量分別為10.39%、9.71%、9.32%、9.51%、9.75%、9.93%、10.35%,健康對照雞為11.05%;蛋雞感染NDV后1~5天,輸卵管組織中膨大部OVA基因mRNA表達(dá)量持續(xù)下降,感染第5天其表達(dá)量僅為健康對照雞的0.08倍;感染后7~15天,感染雞輸卵管膨大部OVA基因mRNA表達(dá)量逐漸回升,感染后第15天,OVA基因mRNA的表達(dá)量達(dá)到健康對照雞的0.27倍。研究表明,NDV感染蛋雞后,感染雞所產(chǎn)雞蛋蛋殼鈣含量的變化與蛋殼質(zhì)量、雞輸卵管子宮部CaBP-D28k基因mRNA的表達(dá)量呈正相關(guān);雞蛋蛋清蛋白含量變化與雞輸卵管膨大部OVA基因mRNA表達(dá)量也呈正相關(guān)。
[Abstract]:Newcastle disease (NDV) is an acute and highly infectious disease of poultry caused by Newcastle disease virus (NDV). Soft shell eggs and deformed eggs, etc. However, the mechanism of fallopian tube pathological changes and egg quality decline after NDV infection in laying hens is unclear. In this study, NDV/Chicken/TC/1/2011 strain was infected with 240-day-old non-immune healthy Roman-white laying hens by nasal drip and eye drops. The egg shell quality of healthy control and infected chickens was detected by collecting eggs produced by healthy control chickens and infected chickens on day 1, 3, 5, 7, 7, 1, 1115 days after infection. The content of calcium in egg shell was detected by atomic spectrophotometry, the ultrastructure of inside, outside and cross section of egg shell was observed by scanning electron microscope, and a certain number of chickens were culled in the healthy control group and the first day after infection. The changes of mRNA expression of CaBP-D28k gene associated with eggshell calcification and egg white quality related ovalbumin (OVA) gene were detected by real-time fluorescence quantitative PCR. The results were as follows: 1. The average relative content (mass ratio) of calcium in eggs produced by infected hens was 40.60,38.41mg / kg, 38.41mg / kg and 38.58.15mg / kg, respectively. The papillomastoid layer thinned, the flatness and compactness of palisade layer and vertical crystal layer decreased in varying degrees, the cuticle on the outer surface of eggshell became rough, stomata enlarged and chapped obviously. The fiber gap on the inner surface of the eggshell gradually increases, the structure is sparse, the fracture is obvious, and the nodes on the eggshell are obviously reduced. The egg shell quality of laying hens improved slowly from 7 to 15 days after infection with NDV. The mRNA expression of CaBPD28k gene in the oviduct and uterus of the laying hens was decreased continuously on the 1st and 5th day after infection with NDV, and the mRNA expression of CaBPD28k gene was only 0.16 times of that of the healthy chickens on the 5th day after infection, and the expression level of CaBPD28k gene was only 0.16 times that of the healthy chickens on the 5th day after infection, and the expression level was 715 days after infection. The quality of egg shell was improved and the expression of CaBP-D28k gene mRNA in uterus was gradually increased. On the 15th day after infection, the expression of CaBP-D28k mRNA in uterus was 0.45 times as high as that in healthy control. The relative contents of protein in egg white of infected hens were 10.39 ~ 9.71and 9.32 ~ 9.75 ~ 9.75 ~ 10.35 days after infection with NDV and 11.05 ~ 15 days after NDV infection in laying hens, the expression of OVA gene mRNA in oviduct tissues of laying hens continued to decrease, and the expression of OVA gene mRNA in oviducts of oviduct tissues continued to decrease, while that of healthy control chickens was 11.05%, and the expression of OVA gene in the oviducts of oviducts continued to decrease after 15 days of infection, while that of healthy control chickens was 11.05%. On the 5th day of infection, the expression level of OVA gene was only 0.08 times of that of the healthy control, and the expression of OVA gene mRNA increased gradually on the 7th and 15th day after infection, and on the 15th day after infection, the expression of mRNA was 0.27 times higher than that of the healthy control. The results showed that there was a positive correlation between the changes of egg shell calcium content and egg shell quality and the expression of CaBP-D28k gene mRNA in oviduct uterus. The change of egg white protein content was also positively correlated with the expression of OVA gene mRNA in the enlarged oviduct.
