【摘要】：[目的]本文旨在確定應(yīng)激對母豬排卵后黃體組織的正常發(fā)育及其功能的影響。[方法]采用注射促腎上腺皮質(zhì)激素(ACTH)的方式模擬應(yīng)激狀態(tài),利用RT-q PCR和免疫組織化學(xué)法研究新生黃體的類固醇合成能力和組織發(fā)育狀況。[結(jié)果]ACTH組母豬血漿中皮質(zhì)醇含量明顯高于對照組母豬,新生黃體內(nèi)膽固醇側(cè)鏈裂解酶P450(P450scc)和3β-羥基類固醇脫氫酶(3β-HSD)的表達(dá)量比對照組母豬明顯下調(diào);ACTH組母豬基質(zhì)金屬蛋白(MMP)和血管內(nèi)皮生長因子(VEGFa)的表達(dá)量明顯低于對照組母豬;組織學(xué)檢測結(jié)果也顯示ACTH組母豬的黃體組織填充率低于對照組母豬,而且黃體內(nèi)血管密度相對較低。[結(jié)論]注射ACTH模擬應(yīng)激,可誘導(dǎo)受試母豬血漿高皮質(zhì)醇含量,而且能夠顯著降低新生黃體內(nèi)主要孕酮合成酶的表達(dá)和黃體組織內(nèi)部的血管新生。提示,應(yīng)激能夠影響母豬新生黃體的正常發(fā)育和功能。
[Abstract]:Objective: to determine the effect of stress on the normal development and function of luteal tissue after ovulation in sows. [methods] the stress state was simulated by injecting adrenocorticotropic hormone (ACTH), and the steroid synthesis ability and tissue development of the corpus luteum were studied by RT-q PCR and immunohistochemistry. [results] the content of cortisol in plasma of ACTH group was significantly higher than that of control group. The expression of cholesterol side chain lyase P450 SCC and 3 尾 -hydroxysteroid dehydrogenase 3 尾 -HSDin in newborn yellow was significantly lower than that in control sows. The expression of matrix metalloprotein (MMPP) and vascular endothelial growth factor (VEGF) in sows of ACTH group was significantly lower than that in control sows. Histological examination also showed that the filling rate of luteal tissue in ACTH sows was lower than that in control sows, and the vessel density in yellow sows was relatively low. [conclusion] injection of ACTH to mimic stress can induce high cortisol content in plasma, and can significantly reduce the expression of progesterone synthase and angiogenesis in corpus luteum. The results suggest that stress can affect the normal development and function of newborn corpus luteum in sows.
