日本血吸蟲(chóng)小RNA let-7對(duì)蟲(chóng)體生殖發(fā)育影響的研究

【摘要】:為檢測(cè)日本血吸蟲(chóng)小RNA let-7(Sja-let-7)對(duì)蟲(chóng)體生殖發(fā)育的影響,本研究在日本血吸蟲(chóng)尾蚴感染BALB/c小鼠后的11 d~42 d,將Sja-let-7的同系物(Sja-let-7 mimics)注射入宿主體內(nèi)對(duì)血吸蟲(chóng)Sja-let-7的表達(dá)進(jìn)行干擾,結(jié)果顯示Sja-let-7 mimics干擾組小鼠肝臟蟲(chóng)卵孵化率顯著升高;小鼠肝臟肉芽腫病變減輕十分明顯,病灶周?chē)仔约?xì)胞浸潤(rùn)半徑變小,空泡變性較對(duì)照組輕微,周?chē)=M織完整性也優(yōu)于對(duì)照組。掃描電鏡和透射電鏡未觀察到蟲(chóng)體的明顯變化。利用熒光定量PCR檢測(cè)Sja-let-7的候選靶基因干擾蟲(chóng)體的表達(dá)情況,結(jié)果表明Sja-let-7 mimics對(duì)大部分候選基因存在不同程度的抑制作用。本研究為后續(xù)深入研究小RNA Sja-let-7的功能奠定了基礎(chǔ)。
[Abstract]:In order to detect the effect of small RNA let-7 Sja-let-7) on the reproductive development of Schistosoma japonicum, the Sja-let-7 congener Sja-let-7 mimicswas injected into the host to interfere with the expression of Sja-let-7 in mice infected with Schistosoma japonicum cercariae at 11 days and 42 days after infection. The results showed that the hatching rate of liver eggs in Sja-let-7 mimics interference group was significantly higher than that in control group, the pathological changes of liver granuloma in mice were obviously alleviated, the infiltration radius of inflammatory cells around the lesion was smaller, and the vacuolar degeneration was less than that in control group. The peripheral normal tissue integrity was also superior to that of the control group. No obvious changes were observed under scanning electron microscope and transmission electron microscope. Fluorescence quantitative PCR was used to detect the expression of candidate target genes in Sja-let-7. The results showed that Sja-let-7 mimics had different inhibitory effects on most candidate genes. This study lays a foundation for further research on the function of small RNA Sja-let-7.
