不同雞品種BNK基因序列比較分析

【摘要】:BNK(C-type lectin-like receptor 2)基因為C型凝結素受體基因中的抑制性基因,與禽類先天免疫性能關系密切。本研究以NCBI中Gallus gallus BNK基因序列為參考,利用MEGA6軟件對24個雞品種的BNK基因進行序列比較分析,篩選出外顯子和內含子上的SNPs位點、Indels以及氨基酸變化,并對24個雞品種進行系統(tǒng)聚類。研究發(fā)現(xiàn),SNPs位點大多發(fā)生在Exon3、Exon5和Exon6上,且多為錯義突變。內含子上的SNPs位點大多發(fā)生在Intron1、Intron2和Intron4上。插入缺失發(fā)生在Intron2、Intron4和Exon5上,并且大多數(shù)品種Indels序列基本一致,國內外雞品種之間并無顯著差異。系統(tǒng)聚類分析將這些雞品種分為3類,狼山雞、羅斯雞、隱性白羽肉雞、茶花雞和如皋黃雞為一類;其次,雪山雞、安卡雞、斗雞和蕭山雞聚為一類;固始雞單獨為一類,表明固始雞的BNK基因為單獨起源。綜上表明,BNK基因在我國地方雞品種中具有豐富的遺傳多樣性,與不同雞品種的先天免疫性能存在關聯(lián),這為家禽的免疫遺傳學和抗病育種提供了參考依據。
[Abstract]:BNK(C-type lectin-like receptor 2) gene is an inhibitory gene of type C lectin receptor gene, which is closely related to the innate immunity of poultry. In this study, the sequence of Gallus gallus BNK gene in NCBI was compared and analyzed by MEGA6 software, and the SNPs loci on exon and intron were screened out. Systematic clustering was carried out on 24 chicken breeds. It was found that most of the SNPs loci occurred on Exon3 exon5 and Exon6, and most of them were missense mutations. Most of the SNPs loci in intron are on Intron1, Intron2 and Intron4. The insertion deletion occurred on Intron2, Intron4 and Exon5, and the Indels sequence of most breeds was basically the same, but there was no significant difference between domestic and foreign chicken breeds. According to the systematic cluster analysis, these chicken breeds were divided into three categories: Wolfhill chicken, Roth chicken, recessive white feather broiler, camellia chicken and Rugao yellow chicken; secondly, Xueshan chicken, Anka chicken, fighting chicken and Xiaoshan chicken were grouped into three categories; The results showed that the BNK base of Gushi chicken was isolated. It is concluded that the BNK gene has abundant genetic diversity in local chicken breeds in China, which is related to the innate immune performance of different chicken breeds, which provides a reference for the breeding of poultry immune genetics and disease resistance.
