峨眉雪膽微丸的制備及對(duì)斷奶仔豬免疫功能和抗氧化性能的影響

【摘要】:養(yǎng)豬行業(yè)長期依靠抗生素來控制仔豬斷奶應(yīng)激綜合征,但是抗生素會(huì)引起仔豬腸道微生物菌群失調(diào)和腸道發(fā)育受損等危害。與在飼糧中添加抗生素相比,添加中草藥提取物,能有效地調(diào)節(jié)斷奶仔豬消化道微生物體系的平衡,促進(jìn)仔豬腸道發(fā)育,從而有效降低腹瀉率,減少斷奶應(yīng)激反應(yīng),有助于提高仔豬的免疫力和生產(chǎn)性能。峨眉雪膽對(duì)腹瀉的治療效果較好,但在養(yǎng)豬生產(chǎn)上的研究還未見報(bào)道。因此,本研究探索了峨眉雪膽作為飼料添加劑對(duì)仔豬斷奶應(yīng)激綜合征的預(yù)防效果。因峨眉雪膽味極苦,直接在飼料中添加會(huì)影響仔豬的適口性,因而將其制備成微丸。同時(shí),研究了峨眉雪膽微丸對(duì)斷奶仔豬免疫功能、血清生化指標(biāo)和抗氧化活性的影響。結(jié)果如下:(1)峨眉雪膽微丸的制備:采用擠出滾圓一流化床包衣法對(duì)其處方條件和工藝條件進(jìn)行考察,主要輔料為微晶纖維素(MCC),擠出轉(zhuǎn)速為40 r/min,滾圓轉(zhuǎn)速為400r/min,滾圓時(shí)間為30s,干燥后經(jīng)流化床包衣制得峨眉雪膽微丸(1 g微丸含原雪膽生藥1 g),圓整度、流動(dòng)性、崩解時(shí)限均較理想,其工藝簡(jiǎn)便,成品質(zhì)量好。(2)峨眉雪膽微丸對(duì)斷奶仔豬免疫功能的影響:與抗生素組相比,斷奶仔豬日糧中添加0.025%峨眉雪膽微丸使斷奶仔豬血清免疫球蛋白IgG、IgM、IgA的含量(P0.05)顯著提高,補(bǔ)體C3、C4的生成量增加以及細(xì)胞因子IL-6的分泌量增多(P0.05),而細(xì)胞因子IL-1β含量顯著降低(P0.05)。并且能顯著提高十二指腸、回腸、盲腸的分泌型免疫球蛋白A(SIgA)的分泌(P0.05)。結(jié)果表明:日糧中添加0.025%峨眉雪膽微丸可增強(qiáng)斷奶仔豬的免疫功能。(3)峨眉雪膽微丸對(duì)斷奶仔豬血清生化指標(biāo)的影響:與抗生素組相比,斷奶仔豬日糧中添加0.025%峨眉雪膽微丸能夠使仔豬血清中總蛋白(TP),白蛋白(ALB)和球蛋白(GLB)的濃度增大,二者在第三周差異顯著。血清中尿素氮的濃度顯著降低。結(jié)果表明:峨眉雪膽微丸可改善斷奶仔豬血清生化指標(biāo)。(4)峨眉雪膽微丸對(duì)斷奶仔豬抗氧化性能的影響:0.025%峨眉雪膽微丸能夠顯著提高血清SOD活性(P0.05),且影響持續(xù)時(shí)間長;能夠提高仔豬GSH活性;在一定程度上降低MDA含量。結(jié)果表明:峨眉雪膽微丸作為中草藥飼料添加劑能夠增強(qiáng)斷奶仔豬的抗氧化能力。
[Abstract]:For a long time, the pig industry relies on antibiotics to control weaning stress syndrome in piglets, but antibiotics can cause intestinal microflora imbalance and intestinal development damage in piglets. Compared with the supplement of antibiotics in diet, the addition of Chinese herbal medicine extract can effectively regulate the balance of digestive tract microbial system, promote the development of intestinal tract of weaning piglets, thus effectively reduce the diarrhea rate and stress response of weaning piglets. It is helpful to improve the immunity and production performance of piglets. Emei Xue gallbladder is effective in treating diarrhea, but the research on pig production has not been reported. Therefore, the preventive effect of Emei Xue gallbladder as a feed additive on weaning stress syndrome in piglets was investigated in this study. Due to the bitter taste of Emei snow gallbladder, adding directly to the feed will affect the palatability of piglets, so it is prepared into pellets. At the same time, the effects of Emei Xuedan pellet on immune function, serum biochemical index and antioxidant activity of weaned piglets were studied. The results were as follows: 1) the preparation of Emei Xuedan pellet: the prescription and technological conditions of the pellet were investigated by extrusion round and fluidized bed coating method. The main excipients were microcrystalline cellulose (MCC), the extrusion speed was 40 r / min, the rotary speed was 400 r / min, and the rolling time was 30 s. After drying and coating with fluidized bed, the Emei snow bile pellet was prepared by fluidized bed coating. The effect of Emei Xuedan pellet on the immune function of weaning piglets was simple and the quality of finished product was good. Compared with the antibiotic group, adding 0.025% Emei Xuedan pellet to the diet of weaning piglets could significantly increase the content of IgGG IgMMA IgA in the serum of weaned piglets (P0.05), compared with the antibiotic group, Emei Xuedan pellet added 0.025% Emei Xuedan pellets to the diet of weaning piglets. The production of complement C _ 3N _ 4 and the secretion of cytokine IL-6 were increased (P 0.05), while the content of cytokine IL-1 尾 decreased significantly (P 0.05). And the secretory immunoglobulin A Siga secretion of duodenum, ileum and cecum was significantly increased (P 0.05). The results showed that the supplementation of 0.025% Emei Xuedan pellet could enhance the immune function of weaned piglets. The effect of Emei Xuedan pellet on serum biochemical indexes of weaned piglets was compared with that of antibiotic group. Adding 0.025% Emei Xuedan pellet to weaning diet could increase the concentrations of total protein (TP), albumin (ALB) and globulin (GLB) in serum of piglets, and the difference was significant in the third week. The concentration of urea nitrogen in serum decreased significantly. The results showed that Emei Xuedan pellet could improve the serum biochemical index of weaning piglets. It can improve the GSH activity of piglets and reduce the content of MDA to some extent. The results showed that Emei Xuedan pellet as a Chinese herbal feed additive could enhance the antioxidant ability of weaned piglets.
