【摘要】：本研究旨在探究不同年齡、不同部位新疆驢皮中Ⅰ型膠原蛋白2個(gè)亞基基因(COL1A1、COL1A2)mRNA的表達(dá)量及其蛋白含量的差異,確定宰驢取皮的年齡段,以提高驢皮的利用率。對(duì)不同年齡段新疆驢背部皮膚組織,應(yīng)用RT-qPCR、Western blotting技術(shù)檢測(cè)膠原蛋白基因的表達(dá)量及其蛋白含量;對(duì)新疆驢不同部位皮膚進(jìn)行HE染色切片與RT-qPCR技術(shù)分析。結(jié)果表明:6~7歲年齡段的驢皮中COL1A1、COL1A2基因的表達(dá)量及其蛋白含量顯著高于其他各年齡段(P0.05)。從組織切片上可以看到,驢皮背部與頸部的膠原纖維密集且多,而臉部的疏松且少,腿部和腹部居中;這與其膠原蛋白基因RT-qPCR的結(jié)果相一致。綜上,6~7歲年齡段的新疆驢背部皮膚中,膠原蛋白基因的表達(dá)量及含量最高,故此年齡段可作為宰驢取皮的最佳年齡段。
[Abstract]:The purpose of this study was to investigate the difference of mRNA expression and protein content of two subunit genes of collagen type I in Xinjiang donkey skin of different ages and different parts, and to determine the age of skin removal from donkey slaughtering in order to improve the utilization rate of donkey skin. The expression of collagen gene and its protein content were detected by RT-qPCR- Western blotting in the dorsal skin of Xinjiang donkey at different ages, and HE staining and RT-qPCR technique were used to analyze the different parts of the skin of Xinjiang donkey. The results showed that the expression of COL1A1A2 gene and its protein content in the donkey skin of 7 years old were significantly higher than those of other age groups (P0.05A). It can be seen from tissue section that the collagen fibers in the back and neck of the donkey skin are dense and much, but the face is loose and less, and the legs and abdomen are in the middle, which is consistent with the result of RT-qPCR. The expression and content of collagen gene were the highest in the back skin of Xinjiang donkeys aged 6 to 7 years old, so the age group could be regarded as the best age for the removal of donkey skin.
