地塞米松對(duì)山羊皮質(zhì)醇分泌和瘤胃生物鐘基因表達(dá)的影響

【摘要】:為了研究地塞米松對(duì)山羊皮質(zhì)醇分泌和瘤胃生物鐘基因表達(dá)的影響,試驗(yàn)選用10只裝有永久性瘤胃瘺管的波爾雜交山羊,隨機(jī)分為對(duì)照組(n=5)和地塞米松組(n=5),對(duì)照組注射生理鹽水,地塞米松組注射地塞米松(0.2 mg/kg),每天注射1次,連續(xù)注射21 d。試驗(yàn)結(jié)束后,采集血液和瘤胃組織;放射免疫分析法(radioimmunoassay,RIA)檢測(cè)血漿皮質(zhì)醇含量;Real-time PCR檢測(cè)瘤胃生物鐘基因表達(dá)量。結(jié)果顯示:與對(duì)照組相比,地塞米松組血漿皮質(zhì)醇濃度顯著降低(P0.05),瘤胃液中皮質(zhì)醇濃度無顯著變化(P0.05),瘤胃上皮組織中永恒蛋白基因(Tim)和肌肉芳香烴受體核轉(zhuǎn)運(yùn)樣蛋白1(Bmal1)mRNA表達(dá)顯著上調(diào)(P0.05);相關(guān)性分析結(jié)果顯示:血漿皮質(zhì)醇濃度與瘤胃上皮組織中Tim(R=-0.64,P=0.08)和Bmal1(R=-0.62,P=0.09)mRNA表達(dá)呈負(fù)相關(guān)趨勢(shì),瘤胃中皮質(zhì)醇濃度與瘤胃上皮組織中Bmal1 mRNA表達(dá)呈顯著負(fù)相關(guān)(R=-0.84,P0.05)。結(jié)果提示,外源性地塞米松降低山羊體內(nèi)內(nèi)源性皮質(zhì)醇的生成,上調(diào)瘤胃上皮組織中生物鐘基因的表達(dá),且生物鐘基因表達(dá)水平與血液、瘤胃液中皮質(zhì)醇濃度呈高度負(fù)相關(guān)。
[Abstract]:In order to study the effect of dexamethasone on cortisol secretion and circadian clock gene expression in goat, 10 Boer hybrid goats with permanent rumen fistula were selected. The rats were randomly divided into two groups: control group (n = 5) and dexamethasone group (n = 5). The control group was injected with normal saline and the dexamethasone group was injected with dexamethasone (0.2 mg / kg 路kg ~ (-1) once a day for 21 days. After the experiment, blood and rumen tissue were collected and radioimmunoassay was used to detect the content of cortisol in plasma and real-time PCR to detect the expression of rumen clock gene. The results showed that: compared with the control group, In dexamethasone group, the plasma cortisol concentration decreased significantly, while the concentration of cortisol in rumen fluid did not change significantly (P 0.05), and the expression of Eternal protein gene (timm) and muscle aromatics receptor nuclear transporter (1(Bmal1)mRNA) in rumen epithelium increased significantly (P 0.05). The results of sex analysis showed that there was a negative correlation between the plasma cortisol concentration and the mRNA expression of Timber-0.64P0.08 and Bmal1r-0.62P0.09 in rumen epithelium. There was a significant negative correlation between the concentration of cortisol in rumen and the expression of Bmal1 mRNA in rumen epithelium. The results suggested that exogenous dexamethasone decreased endogenous cortisol production and up-regulated the expression of clock gene in rumen epithelium, and the expression level of biological clock gene was negatively correlated with blood and cortisol concentration in rumen fluid.
