【摘要】：豬(Sus scrofa)的卵母細(xì)胞體外成熟培養(yǎng)以及胚胎體外培養(yǎng)是豬體外受精、體細(xì)胞克隆以及胚胎干細(xì)胞分離的重要基礎(chǔ)。優(yōu)化卵母細(xì)胞以及胚胎的體外培養(yǎng)條件,是提高豬卵母細(xì)胞體外成熟效率和胚胎體外發(fā)育能力的重要方面。本研究比較了分別添加丙氨酰-谷氨酰胺(alanyl-glutamine,Ala-Gln)和谷氨酰胺(glutamine,Gln)的卵母細(xì)胞成熟培養(yǎng)液、胚胎培養(yǎng)液(porcine zygote medium-3,PZM-3)對(duì)豬卵母細(xì)胞成熟、孤雌激活胚胎和體細(xì)胞克隆胚胎發(fā)育的影響。實(shí)驗(yàn)結(jié)果表明,丙氨酰-谷氨酰胺對(duì)豬卵母細(xì)胞第一極體可見率無明顯提升作用,但可以提高豬孤雌胚胎的囊胚率和囊胚細(xì)胞數(shù),同時(shí)能顯著提高豬體細(xì)胞克隆胚胎的分裂率和囊胚率。本研究為優(yōu)化胚胎體外培養(yǎng)條件、提高豬體細(xì)胞克隆效率提供了一種新的方法。
[Abstract]:In vitro maturation and in vitro embryo culture of Sus scrofa are the important bases for in vitro fertilization, somatic cloning and isolation of embryonic stem cells. To optimize the culture conditions of oocytes and embryos in vitro is an important aspect to improve the efficiency of in vitro maturation of porcine oocytes and the ability of embryo development in vitro. The effects of alanyl-glutamine (Ala-Gln) and glutamine (glutamine) on the development of porcine zygote medium-3PZM-3 and porcine zygote medium-3PZM-3 on porcine oocytes maturation, parthenogenetic embryos and somatic cloned embryos were compared. The results showed that alanyl-glutamine had no effect on the first polar body visibility of porcine oocytes, but could increase the blastocyst rate and the number of blastocysts in parthenogenetic embryos. At the same time, the cleavage rate and blastocyst rate of porcine somatic cloned embryos were significantly increased. This study provides a new method for optimizing the conditions of embryo culture in vitro and improving the cloning efficiency of porcine somatic cells.
