【摘要】：為探索誘導(dǎo)型腹水綜合征肉雞心臟的病理變化與心肌細(xì)胞增殖之間的關(guān)系,將20日齡白羽肉雞分成對(duì)照組(靜脈注射肝素鈉生理鹽水,n=30)和試驗(yàn)組(靜脈注射含纖維素微粒的肝素鈉生理鹽水,n=90)。自注射纖維素微粒后,記錄腹水綜合征(AS)發(fā)病率,并于肉雞42日齡時(shí)取樣,測(cè)定右心室重量與全心室重量之比(mrv/mtv)、紅細(xì)胞壓積(PCV)以及血清中谷草轉(zhuǎn)氨酶(AST)活性濃度和肌酸激酶(CK)活性濃度;右心室組織常規(guī)蘇木精—伊紅染色,并運(yùn)用實(shí)時(shí)熒光定量PCR測(cè)定右心室組織PCNA和Ki-67的表達(dá)量。結(jié)果,與對(duì)照組相比,試驗(yàn)組肉雞AS發(fā)病率、mrv/mtv值、PCV及血清中AST活性濃度和CK活性濃度顯著增高(P0.01);試驗(yàn)組肉雞的右心室明顯擴(kuò)張,心肌細(xì)胞出現(xiàn)水腫現(xiàn)象,但PCNA和Ki-67的m RNA表達(dá)量無(wú)明顯變化。上述結(jié)果表明,誘導(dǎo)型腹水綜合征肉雞心臟產(chǎn)生了嚴(yán)重的病理變化,但心肌所產(chǎn)生的這種病變與細(xì)胞的增殖無(wú)關(guān)。
[Abstract]:To explore the relationship between cardiac pathological changes and cardiomyocyte proliferation in broilers with induced ascites syndrome. White broilers of 20 days old were divided into two groups: control group (intravenous injection of heparin sodium normal saline) and experimental group (intravenous injection of sodium heparin sodium saline containing cellulose particles). The incidence of ascitic syndrome (ASS) was recorded after injecting cellulose particles and was sampled at 42 days of age in broilers. The ratio of right ventricular weight to whole ventricular weight (RV / W), hematocrit (PCV), aspartate aminotransferase (AST) activity and creatine kinase (CK) activity in serum were measured, and the right ventricular tissue was stained with hematoxylin and eosin. The expression of PCNA and Ki-67 in right ventricular tissue was measured by real-time fluorescence quantitative PCR. The results showed that compared with the control group, the incidence of as in the experimental group was significantly higher than that in the control group, and the AST activity and CK activity concentration in serum were significantly higher than those in the control group, while the right ventricle was dilated and the cardiomyocyte edema was observed in the experimental group. However, the expression of m RNA in PCNA and Ki-67 did not change significantly. These results indicated that the heart of broiler with induced ascites syndrome had serious pathological changes, but the pathological changes of myocardium were not related to cell proliferation.
