【摘要】：為了研究重組蛋白UBC13對RAW264.7小鼠巨噬細(xì)胞炎癥因子mRNA表達(dá)量的影響,采用噻唑藍(lán)(MTT)法檢測UBC13對RAW264.7細(xì)胞活力的影響,qPCR法檢測不同濃度UBC13(20、10、5μg/mL)在不同時間點(diǎn)(3、6、9、12、24h)對RAW264.7細(xì)胞產(chǎn)生的IL-6、IL-1β、TNF-α和COX-2 4種炎癥因子mRNA表達(dá)量的影響。結(jié)果表明,在整個試驗(yàn)時間范圍內(nèi)IL-6、IL-1β、TNF-α和COX-2mRNA表達(dá)量呈現(xiàn)逐步降低的趨勢;與細(xì)胞對照組組相比,在24h時IL-6、IL-1β、TNF-α和COX-2mRNA表達(dá)量顯著降低(P0.05)。說明重組蛋白UBC13作用于RAW264.7細(xì)胞24h時能夠明顯抑制RAW264.7細(xì)胞中IL-6、IL-1β、TNF-α和COX-2的mRNA表達(dá),并且具有藥物劑量依賴性。
[Abstract]:In order to study the effect of recombinant protein UBC13 on the expression of macrophage inflammatory factor mRNA in RAW264.7 mice, The effect of UBC13 on the viability of RAW264.7 cells was determined by using thiazolyl blue UBC13 assay. The effects of different concentrations of UBC1320C20 / 105 渭 g / mL) at different time points on the expression of IL-6, IL-1 尾 -TNF- 偽 and COX-2 four inflammatory cytokines mRNA were detected at different time points. The results showed that the expression of IL-6, IL-1 尾, TNF- 偽 and COX-2mRNA decreased gradually in the whole experimental time range, and the expression of IL-6, IL-1 尾, TNF- 偽 and COX-2mRNA decreased significantly at 24 h compared with the control group (P 0.05). The results showed that the recombinant protein UBC13 could inhibit the mRNA expression of IL-6, IL-1 尾, TNF- 偽 and COX-2 in RAW264.7 cells in a dose-dependent manner.
