新種豬源嗜血支原體gapA基因的克隆及分子進化分析

【摘要】:本實驗室前期的流行病學(xué)調(diào)查中發(fā)現(xiàn)浙江省存在新種豬源嗜血支原體(CMh)的流行。為進一步確認該新種CMh,本研究設(shè)計兼并引物獲得CMh gapA基因部分片段,通過Genome walking的方法獲得了全基因cmh-gapA,大小為1 008 bp�;蛐蛄斜葘Ψ治霰砻鱟mh-gapA與犬M.haemocanis的gapA基因親緣關(guān)系最近,同源性為74.6%。核苷酸序列的系統(tǒng)發(fā)育分析表明CMh與犬、貓的嗜血支原體位于同一分支,與M.suis和M.parvum親緣關(guān)系較遠。本研究為CMh作為新種豬源嗜血支原體提供佐證,并為豬源嗜血支原體GADPH蛋白功能研究和血清學(xué)診斷方法的建立奠定基礎(chǔ)。
[Abstract]:An epidemiological investigation in our laboratory revealed the prevalence of new breed Mycoplasma haemophilus (CMH) in Zhejiang Province. In order to further identify the new species of CMH, a partial fragment of CMh gapA gene was obtained by designing degenerate primers, and the whole gene cmh-gapA was obtained by Genome walking with the size of 1 008 BP. The gene sequence alignment analysis showed that cmh-gapA had the closest relationship with the gapA gene of canine M.haemocanis, and the homology was 74.6. Phylogenetic analysis of nucleotide sequence showed that CMh was located in the same branch with dogs and cats, and was closely related to M.suis and M.parvum. This study provided evidence for CMh as a new breed of Mycoplasma sibiricum, and laid a foundation for the study of the function of GADPH protein and the establishment of serological diagnostic method for Mycoplasma suis.
