阿司匹林丁香酚酯的一代繁殖毒性研究

【摘要】:為了初步評(píng)價(jià)阿司匹林丁香酚酯(AEE)對(duì)親代大鼠的生殖功能和子代早期發(fā)育的影響,將360只Wistar大鼠隨機(jī)分成6組,分別為受試藥物AEE高、中、低劑量組(劑量分別為498.0、124.5和31.1 mg·kg~(-1)·d~(-1)),阿司匹林組(劑量為68.4 mg·kg~(-1)·d~(-1)),丁香酚組(劑量為246.0 mg·kg~(-1)·d~(-1)),空白對(duì)照組(給予常規(guī)維持飼料)。各實(shí)驗(yàn)組均按設(shè)定劑量連續(xù)給藥10周后,測(cè)定親代大鼠主要的臟器系數(shù)、繁殖指數(shù)和生產(chǎn)指標(biāo)以及子代(F1)仔鼠的出生指標(biāo)和生長(zhǎng)指標(biāo)。結(jié)果顯示,與空白對(duì)照組相比,高劑量的AEE影響胎鼠的生長(zhǎng)(P0.05),并且高劑量AEE組大鼠的妊娠率、窩產(chǎn)仔數(shù)、出生活仔率和出生存活率都偏低,其中雄性仔鼠比例偏高;阿司匹林組妊娠大鼠和哺乳期胎鼠生長(zhǎng)緩慢(P0.05);高劑量組AEE親代大鼠的睪丸和附睪系數(shù)偏高(P0.05),阿司匹林和丁香酚組子代大鼠的附睪系數(shù)偏高(P0.05)。結(jié)果表明,在本試驗(yàn)條件下,AEE對(duì)大鼠繁殖毒性的最大無(wú)損害作用劑量為31.1 mg·kg~(-1)·d~(-1),對(duì)大鼠的最小有損害作用劑量為124.5 mg·kg~(-1)·d~(-1)。
[Abstract]:In order to evaluate the effect of aspirin eugenol ester (AEE) on the reproductive function and early development of offspring, 360 Wistar rats were randomly divided into 6 groups. The low dose group (498.0124.5 mg / kg) and the low dose group (31.1 mg / kg 路kg ~ (-1)) were treated with a dose of 498.0124.5 and 31.1 mg / kg / d respectively. The aspirin group (68.4 mg / kg) and the eugenol group (246.0 mg / kg / kg) were treated with dimethylpyridamole (DX) -1, respectively, and the blank control group was given routine maintenance diet. The main organ coefficient, reproduction index and production index, birth index and growth index of the offspring were measured after 10 weeks of continuous administration of the drug in each experimental group. The results showed that compared with the blank control group, high dose of AEE affected the growth of fetal rats (P 0.05), and the pregnancy rate, litter size, litter size, survival rate and birth survival rate of the rats in the high dose AEE group were lower than those in the control group, in which the proportion of male rats was higher than that of the control group. The testis and epididymis coefficients of AEE parental rats in aspirin group and eugenol group were higher than that of control group, and the epididymal coefficients of offspring rats in aspirin and eugenol groups were higher than that in control group (P 0.05). The results showed that the maximum noninvasive dose of AEE on reproductive toxicity was 31.1 mg 路kg ~ (-1) 路d ~ (-1) D ~ (-1), and the minimum dose was 124.5 mg 路kg ~ (-1) 路d ~ (-1) 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) 路L ~ (-1) ~ (-1) 路L ~ (-1) ~ (-1) 路L ~ (-1) ~ (-1) ~ (-1).
