ERK信號在玉米赤霉烯酮誘導(dǎo)的睪丸支持細胞自噬中作用的研究

【摘要】:為探討玉米赤霉烯酮(ZEA)雄性生殖毒性作用機理,本試驗以睪丸支持細胞TM4細胞系為對象,研究了ZEA對睪丸支持細胞自噬及相關(guān)信號通路的影響。試驗分別以濃度為0、0.1、1、10、20、30 mol/L的ZEA進行染毒,24 h后,利用CCK-8、激光共聚焦,透射電鏡、Western-blot等技術(shù)檢測了ZEA對TM4細胞的存活率、自噬,MAPK信號通路的影響以及MAPK信號通路與自噬的相互關(guān)系。免疫熒光結(jié)果顯示,隨著ZEA濃度的升高,TM4細胞內(nèi)LC3聚點數(shù)量顯著升高,酸性囊泡的個數(shù)增多;Western-blot結(jié)果顯示,自噬相關(guān)蛋白LC3Ⅱ、Beclin-1、ATG5、ATG7表達顯著升高,而P62蛋白表達顯著下降(P0.05或P0.01);MAPK信號通路中ERK1/2磷酸化水平顯著升高(P0.05或P0.01),加入ERK磷酸化抑制劑U0126后,ZEA誘導(dǎo)TM4自噬水平顯著下降(P0.05或P0.01)。結(jié)果表明,在一定濃度范圍內(nèi),玉米赤霉烯酮可以誘導(dǎo)TM4細胞產(chǎn)生自噬,且發(fā)生自噬與ERK信號通路激活有關(guān)。
[Abstract]:In order to investigate the mechanism of male reproductive toxicity of Zea, the effects of ZEA on testicular Sertoli cell (TM4) cell line were studied. The survival rate of TM4 cells, the effect of autophagy MAPK signaling pathway and the relationship between MAPK signal pathway and autophagy were detected by CCK-8, laser confocal and transmission electron microscopy Western-blot after exposure to ZEA at the concentration of 0 ~ 0.1 ~ 1 ~ 10 ~ 20 ~ 30 mol/L for 24 h respectively. The results of immunofluorescence showed that with the increase of ZEA concentration, the number of LC3 aggregates in TM4 cells increased significantly, and the number of acidic vesicles increased.
