豬7種腹瀉相關(guān)病毒的臨床檢測及豬嵴病毒的遺傳進化分析

【摘要】:為了解引起豬腹瀉疫病的病毒性病原種類及感染現(xiàn)狀,本研究首先針對7種報道較少但可引起豬腹瀉的病毒性病原建立了多重RT-PCR檢測方法,包括豬捷申病毒(PTV)、豬薩佩羅病毒(PSV)、豬丁型冠狀病毒(PDCo V)、豬嵴病毒(PKV)、豬札幌病毒(PSa V)、豬星狀病毒(PAst V)和豬環(huán)曲病毒(PTo V)。利用該方法對419份臨床腹瀉糞便樣品進行篩檢,結(jié)果顯示PKV檢出率最高,陽性率達26.98% 45.79%;PKV和其他病原混合感染率達9.52% 18.54%;赑KV的高檢出率及其可能在豬腹瀉疾病中發(fā)揮的作用,本試驗進一步選取3個PKV陽性樣品進行全基因組序列測定及遺傳進化分析。結(jié)果表明,這3個陽性樣品PKV均歸屬于豬嵴病毒屬,且與其他動物嵴病毒遺傳距離較遠,分別標(biāo)記為CM-PKV、JS-PKV和PD-PKV;它們的全基因組同源性為88.1% 89.1%;CM-PKV與2013年報道的中國株JS-02a-CHN/2013同源性最高,而JS-PKV和PD-PKV則與2008年報道的匈牙利株K-30-HUN/2008/HUN同源性最高。表明上海不同地區(qū)豬群中存在的PKV有明顯的遺傳差異,但毒株遺傳特性的差異與其致病力的相關(guān)性需進一步研究。本研究為深入了解PKV的流行現(xiàn)狀及探討其在豬腹瀉疫情中的作用提供了參考。
[Abstract]:In order to understand the types of viral pathogens and the present situation of infection, a multiplex RT-PCR method was established for the detection of seven viral pathogens which are less reported but can cause pig diarrhea. These include PTV, PTV, PDCo, PKV, PSA, PST, PTO and PTO. The method was used to screen 419 fecal samples of clinical diarrhea. The results showed that the detection rate of PKV was the highest, the positive rate was 26.98% and the mixed infection rate of PKV and other pathogens was 9.52% and 18.54%. Based on the high detection rate of PKV and its possible role in porcine diarrhea, three PKV positive samples were selected for genome sequencing and genetic evolution analysis. The results showed that all of the three positive samples belonged to the genus porcine cristae, and had a long genetic distance from other animal cristae viruses, which were labeled CM-PKV JS-PKV and PD-PKV, respectively, and had the highest homology with Chinese strain JS-02a-CHN/2013 reported in 2013. JS-PKV and PD-PKV had the highest homology with the Hungarian strain K-30-HUN/2008/HUN reported in 2008. The results indicated that PKV in pig populations in different areas of Shanghai had obvious genetic differences, but the correlation between genetic characteristics of virulent strains and pathogenicity of the strains needed further study. This study provides a reference for further understanding of the prevalence of PKV and its role in swine diarrhea.
