建立柱前衍生化-HPLC法測定發(fā)酵液中L-蛋氨酸

【摘要】:擬建立一種簡單、快速、高效的柱前衍生化HPLC法測定發(fā)酵液中蛋氨酸含量。以2,4-二硝基氯苯(CDNB)為衍生化試劑,采用高效液相色譜法,定量測定發(fā)酵液中的蛋氨酸含量。色譜條件:流動相0.01 mol/L p H 5.2 Na AC-乙腈(70∶30);檢測波長:350 nm;柱溫:25℃;流速:0.8 m L/min。結(jié)果表明:蛋氨酸含量在10~80μg/m L范圍內(nèi)峰面積與濃度呈現(xiàn)良好的線性關(guān)系,R2=0.999 24;檢測限和定量限分別為0.1和0.5μg/m;加樣回收率為98.7%,并且具有良好的重復(fù)性。
