【摘要】：植物乳桿菌作為微生態(tài)飼料添加劑飼喂后易受消化道胃酸、膽汁酸和消化酶等不利因素的影響。泰山松花粉多糖(TPPPS)作為一種水溶性植物多糖也易受到胃酸和酶的破壞。為了保護植物乳桿菌和TPPPS的生物活性,并探索二者之間是否存在協(xié)同作用。本研究以植物乳桿菌和TPPPS為芯材制備微膠囊,通過正交試驗對包埋工藝進行優(yōu)化,并對最佳條件下制備的微膠囊進行性能檢測。選取體重20 g左右無特定病原體(SPF)小鼠48只,隨機分為4組,每組12只。4組小鼠每天分別口服飼喂1 m L的植物乳桿菌、植物乳桿菌+0.5%TPPPS、微膠囊、磷酸鹽緩沖液(PBS)。飼養(yǎng)試驗42 d。每隔1周各組隨機取3只小鼠稱重記錄,剖檢后無菌采集腸道組織,測定小腸絨毛和隱窩發(fā)育差異及乳酸菌和大腸桿菌定植情況。結果顯示:1)微膠囊包埋率達81.6%,粒徑為84.3μm。在模擬胃液中處理120 min植物乳桿菌存活率為62.36%,37℃條件下存儲120 d存活率達45.9%。2)與PBS組相比,植物乳桿菌、植物乳桿菌+0.5%TPPPS、微膠囊均能不同程度促進小鼠體重增加,提高小腸中乳酸菌數(shù)量,降低大腸桿菌數(shù)量,改善小腸絨毛和隱窩發(fā)育,微膠囊效果最優(yōu)。結果提示,植物乳桿菌協(xié)同TPPPS使用的益生效果顯著優(yōu)于單獨的植物乳桿菌,二者制備的微膠囊能夠進一步提高其益生效果。
[Abstract]:Lactobacillus plantarum, as a microecological feed additive, is susceptible to adverse factors, such as gastric acid, bile acid and digestive enzymes.As a kind of water soluble plant polysaccharides, TPPPSs of Pinus tabulaeformis pollen are also easily damaged by gastric acid and enzyme.In order to protect the biological activity of Lactobacillus plantarum and TPPPS, and explore the synergy between them.In this study, microcapsules were prepared with Lactobacillus plantarum and TPPPS as the core material. The embedding process was optimized by orthogonal test, and the properties of the microcapsules prepared under the optimum conditions were tested.Forty-eight mice weighing about 20 g without specific pathogen were randomly divided into 4 groups. Each group of 12 mice were given 1 mL Lactobacillus plantarum, 0.5 TPPPS, microcapsule, and phosphate buffer solution PBSN.Feeding experiment 42 days.Every 1 week, 3 mice were randomly selected for weighing records. The intestinal tissues were collected without bacteria after dissection, and the differences in the development of villus and crypt and the colonization of lactic acid bacteria and Escherichia coli were measured.The results showed that the embedding rate of microcapsules was 81.6 渭 m and the particle size was 84.3 渭 m.The survival rate of Lactobacillus plantarum treated in simulated gastric juice for 120 min was 62.36 鈩，
