【摘要】：為了建立一種高通量、重復(fù)性好、特異性高的豬繁殖與呼吸綜合征病毒(PRRSV)抗體的快速檢測方法,本研究用純化后的PRRSV非結(jié)構(gòu)蛋白7(Nsp7)重組蛋白作為偶聯(lián)抗原建立PRRSV抗體的熒光微球免疫學(xué)檢測方法。對檢測方法進(jìn)行分析結(jié)果表明,批內(nèi)、批間變異系數(shù)分別為3.2%、4.2%,與其他常見豬病陽性血清無交叉反應(yīng)。當(dāng)單抗?jié)舛鹊椭? ng/mL時,該方法仍具有高靈敏度。本研究建立的熒光微球免疫學(xué)檢測方法能快速檢測臨床樣本PRRSV抗體,為PRRSV檢測提供了一種新方法。
[Abstract]:To establish a rapid detection method for PRRSVV antibody of porcine reproductive and respiratory syndrome virus (PRRSVV) with high throughput, good reproducibility and specificity.In this study, a fluorescent microsphere immunological assay for the establishment of PRRSV antibodies using purified PRRSV nonstructural protein 7nsp7) as a conjugated antigen was carried out.The results showed that the coefficient of variation between batches was 3.2 and 4.2, and there was no cross reaction with other common swine disease positive sera.The method still has high sensitivity when the concentration of McAb is as low as 1 ng/mL.The fluorescent microsphere immunoassay developed in this study can quickly detect PRRSV antibodies in clinical samples and provides a new method for PRRSV detection.
