狐源維氏氣單胞菌體內(nèi)誘導(dǎo)基因的篩選

【摘要】:為了篩選狐源維氏氣單胞菌體內(nèi)誘導(dǎo)基因,本研究將狐源維氏氣單胞菌人工感染家兔,并取不同感染時(shí)期的血清混合,分別用體外培養(yǎng)的維氏氣單胞菌和大腸桿菌BL21全菌、二者菌體裂解物以及熱變性裂解物進(jìn)行吸附,用ELISA進(jìn)行檢測(cè);同時(shí)構(gòu)建狐源維氏氣單胞菌基因組的p ET系統(tǒng)重組質(zhì)粒表達(dá)文庫(kù)。再用經(jīng)吸附處理的血清對(duì)狐源維氏氣單胞菌基因組文庫(kù)進(jìn)行菌落原位免疫雜交篩選,將篩選的陽(yáng)性克隆進(jìn)行測(cè)序,并用RT-PCR進(jìn)一步驗(yàn)證。結(jié)果顯示,最終獲得8個(gè)狐源維氏氣單胞菌的體內(nèi)誘導(dǎo)基因,分別為GTP焦磷酸激酶基因、轉(zhuǎn)錄調(diào)控基因Mar R基因、氫化酶細(xì)胞膜亞基基因、外膜受體轉(zhuǎn)運(yùn)蛋白Ton B基因、Ⅲ型分泌系統(tǒng)asc V基因以及3個(gè)保守假想蛋白基因。研究結(jié)果為維氏氣單胞菌保護(hù)性抗原的篩選、分子致病機(jī)制及跨物種間傳播機(jī)制奠定了基礎(chǔ)。
[Abstract]:In order to screen inducing genes of Aeromonas virginii in vivo, the rabbits were artificially infected with Aeromonas vulgare, and the sera of different infection stages were mixed with Aeromonas Vickers and Escherichia coli BL21 in vitro, respectively.They were adsorbed and detected by ELISA, and the recombinant plasmid library of pET system of Aeromonas foxogenes was constructed.The genomic library of Aeromonas vulgaris was screened by in situ hybridization with the adsorbed serum. The positive clones were sequenced and further verified by RT-PCR.The results showed that eight inducing genes of Aeromonas viride were obtained: GTP pyrophosphokinase gene, Mar R gene, hydrogenase cell membrane subunit gene, outer membrane receptor transporter Ton B gene.Type 鈪,
