兩株生物學(xué)特性不同的新城疫病毒La Sota毒株誘導(dǎo)DF-1細(xì)胞天然免疫反應(yīng)的比較

【摘要】:為探討兩株不同生物學(xué)特性的新城疫病毒La Sota毒株(La Sota參考毒株AV1615;La Sota突變株P(guān)2M1)引起的宿主天然免疫反應(yīng)的差異,將兩個(gè)毒株分別感染雞胚成纖維細(xì)胞(DF-1),Real-time PCR及Western blot檢測(cè)TLR3、TLR7、MDA5等受體的mRNA及蛋白表達(dá);ELISA及免疫熒光檢測(cè)細(xì)胞核轉(zhuǎn)錄因子IRF3的含量及其磷酸化水平;蛋白芯片檢測(cè)40種細(xì)胞因子的分泌水平,并用ELISA驗(yàn)證部分與炎癥、抗病毒等相關(guān)的細(xì)胞因子在細(xì)胞上清中的含量差異。結(jié)果顯示,La Sota參考毒株AV1615感染細(xì)胞后,誘導(dǎo)產(chǎn)生的TLR3、TLR7、MDA5的mRNA及蛋白含量顯著高于La Sota突變株P(guān)2M1(P0.05)。La Sota感染細(xì)胞后12 h,IRF3及其磷酸化水平、細(xì)胞上清中大部分細(xì)胞因子(31/40)的表達(dá)高于P2M1感染組。結(jié)果說(shuō)明,兩株La Sota毒株在誘導(dǎo)DF-1細(xì)胞的天然免疫反應(yīng)方面存在差異,這可能是La Sota P2M1在雞胚中更容易增殖的原因。
[Abstract]:To investigate the difference of host innate immune response induced by two different biological characteristics of Newcastle disease virus La Sota strain La Sota reference strain AV1615 La Sota mutant P2M1.Real-time PCR and Western blot were used to detect the expression of mRNA and protein of TLR3 and TLR7MDA5, respectively. The level of nuclear transcription factor IRF3 and its phosphorylation were detected by immunofluorescence.Protein chip was used to detect the secretion level of 40 cytokines, and ELISA was used to verify the difference of cytokines in the supernatant of some cytokines, such as inflammation, antivirus and so on.The results showed that the mRNA and protein contents of TLR3 + TLR7MDA5 induced by AV1615, a reference strain of La Sota, were significantly higher than those of P2M1(P0.05).La Sota, a mutant strain of La Sota, at 12 h after infection, and the level of IRF3 and phosphorylation of TLR3 was significantly higher than that of P2M1(P0.05).La Sota.The expression of most cytokines (31 / 40) in the supernatant was higher than that in the P2M1 infected group.The results showed that there was a difference between the two strains in inducing the innate immune response of DF-1 cells, which may be the reason why La Sota P2M1 proliferates more easily in chicken embryos.
