不同日齡仔豬外周血T淋巴細(xì)胞的檢測與分析

【摘要】:為建立健康仔豬外周血淋巴細(xì)胞亞群的臨床參考范圍,試驗分析不同生長階段仔豬的免疫系統(tǒng)功能水平,分別在健康仔豬斷奶后第1、7、21和35天采取外周血,分離外周血單個核細(xì)胞,采用三色流式細(xì)胞術(shù)的方法檢測各類主要T淋巴細(xì)胞亞群所占外周血單個核細(xì)胞的比例,及其占CD3~+T淋巴細(xì)胞亞群中的比例,計算CD4~+/CD8~+的比值。結(jié)果表明,隨著日齡的增加公母仔豬CD3~+T淋巴細(xì)胞亞群占外周血單個核細(xì)胞的比例逐漸升高,分別從(38.00±10.94)%和(38.17±15.39)%上升至(55.43±12.30)%和(51.57±10.60)%。在斷奶后1 d和7 d公母仔豬CD3~+T淋巴細(xì)胞比例無顯著性差異(P0.05),至斷奶后21 d母仔豬CD3~+T淋巴細(xì)胞比例顯著增加(P0.05),公仔豬至35 d時CD3~+T淋巴細(xì)胞比例顯著增加(P0.05)。隨著日齡的增長,公母仔豬CD4~+T淋巴細(xì)胞亞群占外周血單個核細(xì)胞的比例分別由(10.29±2.71)%和(10.3±2.17)%上升至(22.2±5.87)%和(23.4±6.84)%。公母仔豬CD4~+T淋巴細(xì)胞的比例逐漸升高,且各個時間點公母仔豬結(jié)果無明顯差異(P0.05)。公母仔豬CD8~+T淋巴細(xì)胞占外周血單個核細(xì)胞的比例在各時間點分別為(23.80±6.67)%、(17.14±10.64)%、(25.46±12.32)%、(33.33±8.93)%和(28.55±6.75)%、(23.62±11.51)%、(37.37±13.18)%、(38.31±15.66)%。隨著日齡的增加公母斷奶仔豬CD8~+T淋巴細(xì)胞呈現(xiàn)先下降后升高的趨勢。斷奶仔豬各淋巴細(xì)胞亞群分布會隨著斷奶時間的增加而呈現(xiàn)不同的變化,并且存在著性別上的差異。結(jié)合兩種不同的細(xì)胞圈定方法,能夠更準(zhǔn)確地反映出仔豬體內(nèi)各淋巴細(xì)胞亞群數(shù)量和比例的變化。
[Abstract]:In order to establish the clinical reference range of peripheral blood lymphocyte subsets of healthy piglets, the immune system function of piglets at different growth stages were analyzed, and peripheral blood was taken on the 1st and 35th day after weaning, respectively.Peripheral blood mononuclear cells (PBMC) were isolated. The percentage of major T lymphocyte subsets in peripheral blood mononuclear cells (PBMC) and the percentage of CD3- T lymphocyte subsets (CD3- T lymphocyte subsets) in peripheral blood mononuclear cells (PBMC) were detected by three color flow cytometry. The ratio of CD4 to / / CD8 ~ was calculated.The results showed that the percentage of CD3- T lymphocyte subsets in peripheral blood mononuclear cells (PBMC) of male and female piglets gradually increased with the increase of age, from 38.00 鹵10.94% and 38.17 鹵15.39% to 55.43 鹵12.30% and 51.57 鹵10.60%, respectively.There was no significant difference in the proportion of CD3 ~ T lymphocytes between male and female piglets on day 1 and 7 after weaning. The percentage of CD3 ~ T lymphocytes in female piglets increased significantly at 21 days after weaning, and the proportion of CD3 ~ T lymphocytes in female piglets increased significantly at 35 days after weaning, and the proportion of CD3 ~ T lymphocytes in female piglets increased significantly at 35 days after weaning.The percentage of CD4T lymphocyte subsets in peripheral blood mononuclear cells (PBMC) of male and female piglets increased from 10.29 鹵2.71% and 10.3 鹵2.17% to 22.2 鹵5.87% and 23.4 鹵6.84%, respectively.The percentage of CD4 ~ T lymphocytes in male and female piglets increased gradually, and there was no significant difference in the results of male and female piglets at different time points.The percentage of CD8T lymphocytes in peripheral blood mononuclear cells (PBMC) of male and female piglets was 23.80 鹵6.67g / L at each time point, respectively. The ratio of CD8T lymphocytes to peripheral blood mononuclear cells (PBMC) was 17.14 鹵10.64 and 25.46 鹵12.32 鹵33.33 鹵8.93% and 28.55 鹵6.75 + 23.62 鹵11.511,37.37 鹵13.1838.31 鹵15.66m respectively.With the increase of age, CD8 ~ T lymphocytes of male and female weaned piglets decreased first and then increased.The distribution of lymphocyte subsets in weaned piglets varied with the increase of weaning time, and there were gender differences.Combined with two different methods of cell delineation, the number and proportion of lymphocyte subsets in piglets can be more accurately reflected.
