文章摘要:目的 構(gòu)建絕經(jīng)后骨質(zhì)疏松疾病動物模型,探討該疾病的發(fā)病機(jī)制與防治措施。方法 將3個月齡健康雌性SD大鼠40只,隨機(jī)分為2組,分別為去卵巢組和假手術(shù)組。去卵巢組切除大鼠雙側(cè)卵巢,手術(shù)后3個月通過檢測體重、陰道涂片、血鈣、子宮濕重、骨組織病理學(xué)、骨組織形態(tài)計量參數(shù)等對模型進(jìn)行評價。結(jié)果 手術(shù)后3個月,去卵巢組大鼠體重較假手術(shù)組明顯增加(P〈0.05),子宮濕重、血鈣含量較假手術(shù)組量明顯下降(P〈0.05),陰道涂片顯示去卵巢組大鼠均無發(fā)情表現(xiàn),骨組織病理學(xué)觀察顯示去卵巢大鼠脛骨骨小梁斷裂,小梁間距變寬,結(jié)構(gòu)紊亂,骨形態(tài)計量學(xué)數(shù)據(jù)顯示去卵巢大鼠脛骨、股骨干骺端骨小梁面積百分率、骨小梁平均厚度均較假手術(shù)組減少(P〈0.05)。結(jié)論 3個月齡SD大鼠去卵巢后3個月可成功復(fù)制出絕經(jīng)后骨質(zhì)疏松模型。
Abstr:Purpose To establish an animal model of women' s postmenopausal osteoporosis, and to explore the pathogenesis ; the prevention and control measures of the disease. Methods Forty female rats aged 3 months old were divided into ovariectomized group and sham operation group. Bilateral ovaries were removed in the ovariectomized group. The body weight, vaginal smear, serum calcium, wet weight of uterine, and the bone tissue histopathological and morphometrie analysis data were collected at 3 months after operation. Results At 3 months after operation, the body weight were increased in the ovariectomized group compared with the sham operation group and the difference was statistically significant ( P 〈 0.05 ) , the wet weight of uterine and the serum calcium was decreased in the ovariectomized group compared with the sham operation group ( P 〈 0. 05 ), vaginal smears showed that there were no estrus in the the ovarieetomized group. The hone histopathology results showed that the tihial trahecular fraetured, trabecular spacing widened, and the bone structure was disorder in the ovariectomized group. Morphometfic analysis showed that the area and thickness of the trahecular bone of tibia and femur in the ovariectomized animals were decreased compared with that in the sham operation group ( P 〈 0.05 ). Conclusion The postmenopausal osteoporosis model can he sueessfully established in 3 months old rats 3 months after ovariectomy operation.
