廣西梧州地區(qū)近十年傳染性法氏囊病病毒vVP2變化特征分析

【摘要】:為了解近10年來(lái)廣西梧州地區(qū)雞傳染性法氏囊病病毒(IBDV)分子進(jìn)化情況,對(duì)2013年—2014年間來(lái)自該地區(qū)傳染性法氏囊病(Infectious bursal disease,IBD)的法氏囊樣品進(jìn)行IBDV的分離鑒定,并對(duì)分離株以及課題組2006年—2013年間分離的毒株,共24株的VP2高變區(qū)(vVP2)進(jìn)行序列分析和遺傳進(jìn)化分析。結(jié)果表明,QX0601等23個(gè)分離株在關(guān)鍵氨基酸位點(diǎn)上具有256I、284A、294I等超強(qiáng)毒株(vvIBDV)的分子特征,遺傳進(jìn)化分析表明,這23株分離株與UK661、HK46等超強(qiáng)毒參考株同處一個(gè)分支中,親緣關(guān)系較近;QX110603在關(guān)鍵性氨基酸位點(diǎn)上則具有256V、284T、294L等弱毒株的特征,遺傳進(jìn)化分析顯示,其與BJ836等致弱株處于同一分支,親緣關(guān)系較近。對(duì)所有分離株進(jìn)行氨基酸位點(diǎn)分析發(fā)現(xiàn),該地區(qū)IBDV進(jìn)化出現(xiàn)了新的特點(diǎn),212D-212N符合國(guó)內(nèi)近年來(lái)的分離株的變化趨勢(shì),209T-209A、338R-338H、359T-359R則表現(xiàn)出地域特點(diǎn),未曾見過(guò)相似報(bào)道。研究結(jié)果表明,具有vvIBDV分子特征的分離株是該地區(qū)近10年來(lái)主要流行毒株,該地區(qū)IBDV毒株在vVP2序列上仍處于不斷進(jìn)化中,且?guī)в械赜蛱攸c(diǎn)。
[Abstract]:In order to understand the molecular evolution of infectious bursal disease virus (IBDV) of chickens in Wuzhou, Guangxi, during the period from 2013 to 2014, samples of IBDs from infectious bursal disease (IBD) were isolated and identified by IBDV. The isolates and virulent strains isolated from 2006 to 2013 by our research group were compared with each other. The results of sequence analysis and genetic evolution analysis showed that 23 isolates, including QX0601, had the molecular characteristics of 256IX284Af294I and other super virulent strains (VVIBDVV) at the key amino acid sites, and the genetic evolution analysis showed that, In the same branch of UK-661T HK46 and UK-661N HK46, these 23 strains were closely related to QX110603 at the key amino acid sites, and had the characteristics of 256VX284TTi294L, and the genetic evolution analysis showed that they were in the same branch as BJ836 and other weakening strains. The analysis of amino acid loci of all isolates showed that the evolution of IBDV in this region showed a new characteristic: 212D-212N, which was in line with the trend of variation of isolates in China in recent years (209T-209A, 338R-338Hn359T-359R). No similar reports have been reported. The results show that the isolates with the molecular characteristics of vvIBDV are the main prevalent strains in this region in recent 10 years. The IBDV strains in this region are still evolving in vVP2 sequence and have regional characteristics.
