【摘要】：為建立豬傳染性胃腸炎病毒(TGEV)直觀、特異和快速的檢測方法,將TGEV接種PK-15細胞后,以抗TGEV N蛋白的單克隆抗體為一抗,熒光素FITC標(biāo)記的山羊抗小鼠Ig G為二抗,建立了TGEV的間接免疫熒光(IFA)檢測方法。結(jié)果顯示,TGEV的最佳接種效價為1×102TCID50,培養(yǎng)時間為24 h,細胞最佳固定液為40 m L/L多聚甲醛,最佳封閉條件為10 g/L BSA 37℃封閉30 min,一抗的最佳使用濃度為1 ng/L,二抗的最佳稀釋度為1∶100。對常見的豬流行性腹瀉病毒、豬輪狀病毒和豬圓環(huán)病毒2型等豬病病原檢測均為陰性。結(jié)果表明,建立的檢測方法對TGEV具有良好的特異性。本研究建立的方法為TGEV的實驗室診斷及TGEV在感染細胞中的定位和動態(tài)分布提供了有效的檢測手段。
[Abstract]:In order to establish an intuitive, specific and rapid method for detection of porcine infectious gastroenteritis virus, TGEV was inoculated into PK-15 cells. Monoclonal antibody against TGEV N protein was used as the first antibody and goat anti-mouse IgG labeled with fluorescein FITC as the second antibody. An indirect immunofluorescence assay was established for the detection of TGEV. The results showed that the best titer of TGEV was 1 脳 102 TCID 50, the culture time was 24 h, and the best immobilization solution was 40 mL / L polyformaldehyde.
