【摘要】：為了研究CFL2基因與骨骼肌纖維相關(guān)基因之間的關(guān)系,本試驗(yàn)運(yùn)用已篩選的CFL2低表達(dá)細(xì)胞株,利用實(shí)時(shí)定量和Western blot技術(shù)檢測(cè)CFL2基因低表達(dá)對(duì)肌纖維組成成分MLC、α-actin與肌纖維類型MyHCslow、MyHC2b、MyHC2a、MyHC2x表達(dá)的影響。結(jié)果表明,低表達(dá)CFL2后,肌纖維組成成分MLC和α-actin的表達(dá)均顯著上調(diào)(P0.05)。MyHC2x、MyHC2b、MyHC2a、MyHCslow的表達(dá)均顯著下調(diào)(P0.05)。說明CFL2可能起到通過調(diào)節(jié)MyHC類型轉(zhuǎn)換來達(dá)到調(diào)節(jié)紅白肌肉比例的效果,進(jìn)而直接影響豬肉品質(zhì)。
[Abstract]:In order to study the relationship between CFL2 gene and skeletal muscle fiber related gene, CFL2 low expression cell lines were used in this study. The effects of low expression of CFL2 gene on the expression of myofiber components (MLC, 偽 -actin) and myofiber type MyHCslow-MyHC2bPU MyHC2aH2x were detected by real-time quantitative analysis and Western blot technique. The results showed that the expression of myHC2x was decreased after low expression of MLC, 偽 -actin, and myofibrillar. The expression of MLC and 偽 -actin in muscle fiber components were all significantly up-regulated by P0.05A. MyHC2xC2xPHC2bC2bPHC2bPHC2AH MyHCslow were down-regulated significantly, indicating that CFL2 may play a role in regulating the ratio of red and white muscles by regulating the MyHC type conversion, and then directly affecting the pork quality. 2. The results showed that the expression of P0. 05 and 偽 -actin in myofiber components were significantly increased.
