【摘要】：牛奶是一種含有豐富營(yíng)養(yǎng)物質(zhì)的飲品,也是人體內(nèi)鈣的最佳來(lái)源。隨著我國(guó)奶牛業(yè)的迅猛發(fā)展,牛奶漸漸受到人們的青睞,低脂健康牛奶逐漸成為人們的首選。肉堿棕櫚�；D(zhuǎn)移酶2(Carnitine Palmitoyltransferase II,CPT2)基因是CPT家族中的一個(gè)亞型,CPT2蛋白位于線(xiàn)粒體內(nèi)膜,是線(xiàn)粒體β-氧化的限速酶。CPT2基因來(lái)源于課題組前期針對(duì)高乳脂高蛋白荷斯坦奶牛與低乳脂低蛋白荷斯坦奶牛轉(zhuǎn)錄組學(xué)分析的基礎(chǔ)上,篩選出的差異基因。本研究構(gòu)建了CPT2基因sh RNA干擾載體和過(guò)表達(dá)載體,通過(guò)瞬時(shí)轉(zhuǎn)染法轉(zhuǎn)入到奶牛的乳腺上皮細(xì)胞(Mammary Epithelial Cells,MEC)中,同時(shí)進(jìn)行了m RNA水平和蛋白水平的驗(yàn)證,并測(cè)定了細(xì)胞內(nèi)甘油三酯的含量和脂肪酸含量的變化。結(jié)果表明,干擾CPT2基因后,奶牛乳腺上皮細(xì)胞內(nèi)甘油三酯含量與對(duì)照組相比顯著性升高(p=0.01930.05);CPT2基因表達(dá)水平與長(zhǎng)鏈脂肪酸含量正相關(guān),干擾組與對(duì)照組相比脂肪酸含量顯著降低(p=0.02470.05),過(guò)表達(dá)組與對(duì)照組相比脂肪酸含量顯著性升高(p=0.04470.05)。通過(guò)本研究,我們明確了CPT2基因的對(duì)脂質(zhì)代謝的影響,為日后構(gòu)建特殊群體中國(guó)荷斯坦奶牛奠定了基礎(chǔ)。
[Abstract]:Milk is a rich nutrient drinks, is the best source of calcium in the body. Along with the rapid development of China's dairy industry, milk gradually favored, low-fat milk has gradually become the first choice for people. Carnitine palmitoyl transferase (Carnitine Palmitoyltransferase 2 II, CPT2) gene is a subtype of the CPT family the CPT2 protein located in the inner mitochondrial membrane, is the rate limiting enzyme of.CPT2 gene from mitochondrial beta oxidation in ourlaboratory against high fat high protein and low fat low Holstein Holstein group protein transcription analysis, differential gene screening. This study constructed RNA interference vector of CPT2 gene and sh expression vector into cow mammary epithelial cells by transient transfection method (Mammary Epithelial Cells, MEC), and verify the m RNA and protein level, and test The change of intracellular triglyceride content and fatty acid content. The results show that the interference of CPT2 gene, triglyceride content in bovine mammary epithelial cells compared with the control group increased significantly (p=0.01930.05); the expression level of CPT2 gene with long chain fatty acid content is related to the interference group compared with the control group, the fatty acid content decreased significantly (p=0.02470.05), over expression group compared with the control group, the fatty acid content increased significantly (p=0.04470.05). Through this study, we identified the CPT2 gene effect on lipid metabolism, lays the foundation for constructing the special group of Chinese Holstein day.
