羧甲基纖維素鈉對豬精液常溫保存效果的影響

【摘要】:在豬人工授精技術(shù)中,良好的稀釋劑是提高豬精子質(zhì)量的關(guān)鍵因素之一。為探討羧甲基纖維素鈉(CMC)對豬精液常溫保存效果的影響,本研究以傳統(tǒng)的豬精液保存稀釋劑Zorlesco(-BSA)為基礎(chǔ)稀釋液,通過添加不同濃度的CMC(0、8、16和24μmol/L)對原精液進行稀釋和保存。采用計算機輔助精液分析儀(CASA)檢測精子活率和活力,低滲腫脹法(HOST)檢測精子質(zhì)膜完整性,考馬斯亮藍染色檢測精子頂體完整性。結(jié)果顯示,在豬精液保存的前3d內(nèi),添加不同濃度CMC組的精子活率和活力與對照組相比較高,但差異不顯著(P0.05);保存4~7d時,與對照組相比添加16μmol/L CMC組的精子活率和活力明顯升高(P0.05)。在保存的前4d內(nèi),添加不同濃度CMC組的精子質(zhì)膜完整率和頂體完整率與對照組相比差異不顯著(P0.05),但在保存至5~7d時,添加16μmol/L CMC組的質(zhì)膜完整率和頂體完整率顯著高于對照組(P0.05)。豬精子活力、活率、質(zhì)膜完整率、頂體完整率之間存在極顯著的正相關(guān)關(guān)系(P0.01),在Zorlesco稀釋劑中添加8~24μmol/L的CMC能顯著改善和提高豬精子活率、活力、質(zhì)膜完整性以及頂體完整率,其最佳添加濃度為16μmol/L。CMC可作為精液稀釋劑中的懸浮劑,用于減緩精子沉降速率,提高豬精液保存質(zhì)量。
[Abstract]:In pig artificial insemination, good diluent is one of the key factors to improve the quality of pig sperm. In this study, based on the traditional porcine semen preservation diluent Zorlesco-BSA, the original semen was diluted and preserved by adding different concentrations of CMC0, 816 and 24 渭 mol / L, and the sperm motility and motility were measured by computer aided semen analyzer (CASAs). The sperm plasma membrane integrity was detected by hypotonic swelling method and acrosome integrity was detected by Coomassie brilliant blue staining. The results showed that the sperm motility and motility of porcine semen with different concentrations of CMC were higher than those of the control group within 3 days before the preservation of porcine semen. However, the difference was not significant (P 0.05), the sperm motility and motility of 16 渭 mol/L CMC group were significantly higher than that of the control group at 4 ~ 7 days after storage, and within 4 days before preservation, the sperm motility and motility of the group were significantly higher than that of the control group. There was no significant difference in plasma membrane integrity and acrosome integrity rate between different concentrations of CMC and control group, but the plasma membrane integrity and acrosome integrity rate of 16 渭 mol/L CMC group was significantly higher than that of control group P0.050.The sperm motility and motility of pig spermatozoa were significantly higher than that of control group. There was a significant positive correlation between plasma membrane integrity and acrosomal integrity. The addition of 8 渭 mol/L CMC to Zorlesco diluent could significantly improve the sperm motility, motility, plasma membrane integrity and acrosome integrity of porcine spermatozoa. The best concentration of 16 渭 mol/L.CMC can be used as suspension in semen diluent, which can be used to slow down the rate of sperm sedimentation and improve the quality of porcine semen preservation.
