大腸桿菌耐藥性及氟苯尼考耐藥基因floR的研究

【摘要】:本實驗對某地養(yǎng)殖場豬源和雞源大腸桿菌的耐藥性及氟苯尼考耐藥基因flo R進行分析,旨在了解我國大腸桿菌的耐藥現(xiàn)狀,以降低對養(yǎng)殖業(yè)的危害,減少菌株耐藥性的傳播,為獸醫(yī)臨床用藥及獸藥管理提供科學依據(jù),為新型抗菌藥物的研制奠定理論基礎。采用國標法分離鑒定大腸桿菌,肉湯微量稀釋法進行藥敏實驗,PCR法檢測氟苯尼考flo R基因,膜過濾法進行轉化接合實驗。結果表明:共分離大腸桿菌293株,分離率為73.3%,其中豬源157株,雞源136株;大腸桿菌對磺胺類、四環(huán)素類和氨基糖苷類藥物的耐藥性相對嚴重,對氯霉素類、氟喹諾酮類和β-內(nèi)酰胺類藥物相對敏感;與歐盟EUCAST標準相比,該地區(qū)大腸桿菌的MIC分布出現(xiàn)明顯右移現(xiàn)象;80%以上的大腸桿菌均為多重耐藥菌株,以8重和9重耐藥菌株為主;氟苯尼考耐藥基因flo R攜帶率為61.2%,flo R基因可以在菌株間相互轉移。樣品采集地區(qū)養(yǎng)殖場大腸桿菌污染嚴重,耐藥種類逐漸增多,多重耐藥現(xiàn)象嚴重,質粒介導的耐藥基因可以在菌株間相互轉移。
[Abstract]:In this experiment, the resistance of Escherichia coli and florfenicol resistance gene flo R from pig and chicken were analyzed in order to understand the current situation of drug resistance of Escherichia coli in China, in order to reduce the harm to the breeding industry and reduce the spread of drug resistance of strains. To provide scientific basis for veterinary clinical drug use and veterinary drug management, and to lay a theoretical foundation for the development of new antimicrobial agents, Escherichia coli was isolated and identified by national standard method, and florfenicol flo R gene was detected by broth microdilution assay. The results showed that 293 strains of Escherichia coli were isolated, and the isolation rate was 73.3%, of which 157 strains were from pigs and 136 strains from chicken, and the resistance of Escherichia coli to sulfonamides, tetracyclines and aminoglycosides was relatively serious. Compared with the European Union EUCAST standard, the MIC distribution of Escherichia coli in this area showed obvious right shift phenomenon, more than 80% of the Escherichia coli were multidrug resistant strains, which were sensitive to chloramphenicol, fluoroquinolones and 尾 -lactams. The flo R carrying rate of florfenicol resistance gene was 61.2% and that of floro R gene could be transferred from strain to strain. The contamination of Escherichia coli in culture farm was serious in the area of sample collection, and the kinds of drug resistance increased gradually. Multidrug resistance is serious and plasmid-mediated resistance genes can be transferred from strain to strain.
