口服黃芪多糖增強(qiáng)腸道黏膜免疫及其對(duì)口蹄疫疫苗免疫的影響

【摘要】:將40只小鼠隨機(jī)分為2組,一組小鼠灌服黃芪多糖水溶液,另一組灌服生理鹽水為對(duì)照組,給藥后每周從各組隨機(jī)取5只小鼠,分離脾淋巴細(xì)胞進(jìn)行淋巴細(xì)胞增殖試驗(yàn),并觀察十二指腸黏膜上皮內(nèi)淋巴細(xì)胞(IELs)以及上皮固有層的IgA+細(xì)胞數(shù);將24只小鼠隨機(jī)分為4組,各小鼠分2次,間隔2周注射口蹄疫(FMD)疫苗,其中1~3組的小鼠在疫苗免疫前灌服不同劑量的黃芪多糖,第4組小鼠灌服生理鹽水作為對(duì)照。二免后1~5周,每周采血,檢測(cè)血清IgG及其亞類。結(jié)果表明,小鼠口服黃芪多糖能顯著促進(jìn)小鼠淋巴細(xì)胞對(duì)刀豆蛋白A(ConA)和脂多糖(LPS)的刺激反應(yīng),并增加十二指腸IELs和固有層IgA+細(xì)胞數(shù)量;口服黃芪多糖后接種FMD疫苗,能夠顯著增強(qiáng)血清FMD特異性IgG及其亞類水平。
[Abstract]:Forty mice were randomly divided into two groups: one group was treated with astragalus polysaccharide solution and the other group was treated with normal saline as control group. Five mice were randomly selected from each group every week after administration, and spleen lymphocytes were isolated for lymphocyte proliferation test. The number of IgA cells in intraepithelial lymphocytes and lamina propria of duodenal mucosa were observed, 24 mice were randomly divided into 4 groups, each mice were divided into 2 groups and injected with foot-and-mouth disease vaccine at intervals of 2 weeks. The mice in group 1 and group 3 received different doses of astragalus polysaccharide before immunization, and mice in group 4 were treated with normal saline as control. Blood samples were collected for 5 weeks after the second immunization. Serum IgG and its subclasses were detected. Astragalus polysaccharides could significantly promote the stimulation of lymphocytes to concanavalin Cona and lipopolysaccharide (LPS), and increase the number of duodenal IELs and lamina propria IgA cells. Astragalus polysaccharide was given to FMD vaccine after oral administration of Astragalus polysaccharide. Serum FMD specific IgG and its subclasses were significantly enhanced.
