一株羊源致病性屎腸球菌的分離鑒定及耐藥性分析

【摘要】:試驗(yàn)從甘肅肅南縣某羊場(chǎng)死亡羊中分離出疑似某球菌菌株XCP,為進(jìn)一步確定該致病菌株及其耐藥性情況,進(jìn)行了病原菌分離、革蘭氏染色、生化試驗(yàn)、16SrRNA PCR擴(kuò)增鑒定、小鼠致病力試驗(yàn)、藥敏試驗(yàn)、毒力基因和耐藥基因擴(kuò)增試驗(yàn)研究。結(jié)果發(fā)現(xiàn),該菌革蘭氏染色陽(yáng)性,高鹽、蜜二糖、蔗糖等生化反應(yīng)陽(yáng)性,VP、馬尿酸鹽、阿拉伯糖等生化反應(yīng)陰性;16SrRNA基因PCR擴(kuò)增后,進(jìn)行BLAST比對(duì),結(jié)果與GenBank中的屎腸球菌16SrRNA基因序列同源性為100%;小鼠致病力試驗(yàn)發(fā)現(xiàn),該菌株具有很強(qiáng)的致病性;該菌株對(duì)β-內(nèi)酰胺類抗生素苯唑西林、青霉素G,喹諾酮類抗生素諾氟沙星、環(huán)丙沙星、左氟沙星,以及四環(huán)素等高度耐藥,對(duì)紅霉素、萬(wàn)古霉素、克林霉素等抗生素敏感;毒力基因Asal、cylA、acm和耐藥基因TetM、ant(6)-Ⅰ、aac(6′)-aph(2′′)、ermB擴(kuò)增均為陽(yáng)性。結(jié)果表明,該菌為屎腸球菌(Enterococcus faecium),具有較強(qiáng)的致病性和耐藥性,可能與毒力基因和耐藥基因的高陽(yáng)性率有關(guān)。
[Abstract]:In order to further determine the pathogenic strain and its drug resistance, the pathogen was isolated from a sheep farm dead sheep in Sunan County, Gansu Province. The pathogen was isolated, Gram-stained, and biochemical test was used to amplify 16s rRNA PCR. The virulence test, drug sensitivity test, virulence gene and drug resistance gene amplification test in mice were studied. The results showed that the strain was positive for Gram stain, high salt, honey disaccharide, sucrose and so on. After amplification of 16s rRNA gene of Arabose and other biochemical reaction, BLAST was compared with 16s rRNA gene of Enterococcus faecium in GenBank, the result showed that the sequence of 16s rRNA gene of Enterococcus faecium in GenBank was 100%, the pathogenicity test of mice showed that the strain had strong pathogenicity. The strain was sensitive to erythromycin, vancomycin, clindamycin and so on, and was highly resistant to 尾 -lactam antibiotics such as oxacillin, penicillin G, quinolones antibiotics norfloxacin, ciprofloxacin, levofloxacin, and tetracycline.
