【摘要】：為探究Lpin3基因變異對綿羊肉用性狀的影響,選擇廣靈大尾羊和小尾寒羊兩品種脂尾型綿羊為研究對象,采用DNA直接測序法檢測Lpin3基因5'非編碼區(qū)部分序列的單核苷酸多態(tài)性(SNPs),并分析SNPs與尾型和屠宰性狀的關聯(lián)。結果檢測到Lpin3基因5'非編碼區(qū)起始密碼子上游約1 200 bp的DNA序列13個SNPs,其中-334~-332ins TAA顯著降低了小尾寒羊的尾寬(P0.05),提高了小尾寒羊的屠宰性狀(P0.05),在小尾寒羊中的突變頻率顯著高于廣靈大尾羊(P0.05),對廣靈大尾羊各性狀無顯著影響(P0.05)。盡管不顯著,小尾寒羊中-443 TC與-334~-332 ins TAA具有相似的遺傳效應。該研究可為綿羊Lpin3基因功能及肉質性狀分子標記輔助選擇提供依據(jù)。
[Abstract]:In order to investigate the effect of Lpin3 gene variation on meat traits of sheep, two types of fat-tailed sheep, Guangling large tail sheep and small tail Han sheep, were selected as the research objects. The single nucleotide polymorphisms (SNPs) of 5'non-coding region partial sequence of Lpin3 gene were detected by DNA direct sequencing, and the association of SNPs with tail type and slaughter traits was analyzed. The results showed that the 5'noncoding region of Lpin3 gene was about 1,200 upstream of the start codon of 5'non-coding region. The BP DNA sequence was 13 SNPs, of which -334N -332ins TAA significantly reduced the tail width of small tail Han sheep (P0.05N), and increased the slaughter trait P0.05N of small tail Han sheep. The mutation frequency in small tail Han sheep was significantly higher than that in small tail Han sheep (P0.05A), but there was no significant difference among the traits of Guangling large tail sheep. P0.05, although not significant, The genetic effects of -443TC and -334n-332 ins TAA were similar in small tail Han sheep. This study could provide evidence for molecular marker assisted selection of Lpin3 gene function and meat quality traits in sheep.
