響應(yīng)面法對(duì)磷酸化淫羊藿多糖制備工藝的研究

【摘要】:采用混合磷酸鹽法制備磷酸化淫羊藿多糖,再以磷酸化淫羊藿多糖磷酸根含量為考察指標(biāo),通過(guò)響應(yīng)面法對(duì)工藝的提取參數(shù)進(jìn)行優(yōu)化,研究了修飾條件中反應(yīng)溫度(A)、反應(yīng)時(shí)間(B)和反應(yīng)p H值(C)3個(gè)因素對(duì)磷酸化多糖磷酸根含量的影響,建立了回歸模型,驗(yàn)證了其有效性,并分析了主效應(yīng)和因素交互作用。結(jié)果表明:反應(yīng)溫度、反應(yīng)時(shí)間和反應(yīng)p H值對(duì)磷酸化淫羊藿多糖磷酸根含量的影響顯著,并且兩兩因素間存在一定的交互作用;影響磷酸化淫羊藿多糖的工藝因素按主次順序排列為:反應(yīng)溫度反應(yīng)時(shí)間反應(yīng)p H值;磷酸化淫羊藿多糖的最佳工藝為:三聚磷酸鈉與三偏磷酸鈉比例為5∶2,反應(yīng)溫度97℃,反應(yīng)時(shí)間2.8 h,反應(yīng)p H 8.7,在此條件下,磷酸化淫羊藿多糖磷酸根含量為(6.012±0.021)%。
[Abstract]:The phosphorylated epimedium polysaccharide was prepared by mixed phosphate method. The phosphate content of the phosphorylated epimedium polysaccharide was used as the index. The extraction parameters were optimized by response surface method. The effects of three factors, such as reaction temperature, reaction time, reaction time and pH value, on phosphorylated polysaccharide phosphate content were studied. A regression model was established to verify its validity. The main effects and the interaction of factors were analyzed. The results showed that the reaction temperature, reaction time and pH value had significant effects on the phosphorylation of epimedium polysaccharide phosphate content, and there was a certain interaction between two factors. The technological factors affecting the phosphorylation of epimedium polysaccharide were as follows: reaction temperature, reaction time, pH value, the optimum process of phosphorylation of epimedium polysaccharide was: the ratio of sodium tripolyphosphate to sodium trimetaphosphate was 5: 2, and the reaction temperature was 97 鈩,
