鐮刀菌毒素對(duì)斷奶仔豬肝臟和脾臟損傷的影響

【摘要】:本研究通過對(duì)血清肝功能指標(biāo)、肝脾的抗氧化指標(biāo)和炎性因子表達(dá)量的測定,以及對(duì)肝脾組織病理學(xué)的觀察,系統(tǒng)研究鐮刀菌毒素對(duì)斷奶仔豬肝臟和脾臟損傷的影響以及停飼鐮刀菌毒素污染飼糧后仔豬的自然恢復(fù)情況。為研究鐮刀菌毒素對(duì)斷奶仔豬肝臟和脾臟損傷的影響,試驗(yàn)選用35日齡、體重為8.45±0.94 kg的健康三元雜交(杜×長×大)雌性仔豬50頭,隨機(jī)分為2組,對(duì)照組(Control group)20頭,鐮刀菌毒素組(Fusarium toxins group)30頭,每頭豬作為1個(gè)重復(fù),對(duì)照組飼喂基礎(chǔ)飼糧,鐮刀菌毒素組飼喂用霉變玉米和霉變玉米蛋白粉替代基礎(chǔ)飼糧中50%的玉米和玉米蛋白粉配制而成的鐮刀菌毒素污染飼糧(0.90 mg/kg ZEN,1.43 mg/kg DON,5.85 mg/kg FUM),預(yù)試期7 d,正試期35 d。正試期結(jié)束后每組隨機(jī)挑選10頭仔豬進(jìn)行屠宰取樣。結(jié)果表明:(1)與對(duì)照組相比,鐮刀菌毒素組斷奶仔豬血清酶活性顯著升高,肝臟蛋白合成能力和抗氧化能力顯著降低(P0.05),且血清相應(yīng)抗氧化指標(biāo)的變化與肝臟一致;肝臟相對(duì)重量顯著降低,炎性細(xì)胞因子IL-1β和IL-6表達(dá)量顯著升高(P0.05),且分布發(fā)生變化;病理學(xué)觀察發(fā)現(xiàn)肝臟出現(xiàn)肝小葉邊緣出現(xiàn)明顯的炎性細(xì)胞浸潤,局部肝細(xì)胞出現(xiàn)空泡變性。(2)與對(duì)照組相比,鐮刀菌毒素組斷奶仔豬脾臟的相對(duì)重量無明顯變化(P0.05),抗氧化能力顯著降低(P0.05),且與血清和肝臟中相應(yīng)指標(biāo)的變化一致;炎性細(xì)胞因子IL-1β和IL-6表達(dá)量顯著升高(P0.05),且分布發(fā)生變化;病理學(xué)觀察發(fā)現(xiàn)脾臟白髓區(qū)明顯變小,紅髓區(qū)擴(kuò)張,且局部出現(xiàn)空泡變性。為研究停飼鐮刀菌毒素污染飼糧后斷奶仔豬肝臟和脾臟損傷的自然恢復(fù)情況,于第一部分屠宰結(jié)束后,從鐮刀菌毒素組隨機(jī)挑選10頭仔豬為自然恢復(fù)組,停止飼喂鐮刀菌毒素污染飼糧,改飼基礎(chǔ)飼糧,其余仔豬仍在原組,每頭豬為1個(gè)重復(fù),試驗(yàn)期21 d。試驗(yàn)結(jié)束后屠宰所有仔豬并取樣。結(jié)果表明:與鐮刀菌毒素組相比,停飼鐮刀菌毒素污染日糧后,斷奶仔豬血清酶活性顯著降低,肝臟蛋白合成能力得到明顯改善(P0.05),但仍未恢復(fù)至對(duì)照組水平(P0.05);肝臟和脾臟的抗氧化能力均有所提高,且與血清抗氧化指標(biāo)的變化一致;肝臟和脾臟組織炎性細(xì)胞因子IL-1β和IL-6表達(dá)量均顯著降低(P0.05),且與對(duì)照組差異均不顯著(P0.05)。以上研究表明,鐮刀菌毒素(0.90 mg/kg ZEN,1.43 mg/kg DON,5.85 mg/kg FUM)可引起斷奶仔豬血清肝功能指標(biāo)的改變、肝臟和脾臟抗氧化能力的降低以及組織炎性細(xì)胞因子表達(dá)的異常,從而造成肝臟和脾臟的損傷;停飼鐮刀菌毒素污染飼糧后,斷奶仔豬的肝臟和脾臟損傷有明顯恢復(fù),但并不能完全恢復(fù)至正常水平。
[Abstract]:In this study, the indexes of serum liver function, the antioxidant index of liver and spleen and the expression of inflammatory factors were measured, and the histopathology of liver and spleen were observed. The effects of Fusarium toxin on liver and spleen injury in weaning piglets and the natural recovery of piglets after stopping feeding with Fusarium toxin contaminated diet were studied systematically in order to study the effects of Fusarium toxin on liver and spleen injury in weaned piglets. A total of 50 female piglets were randomly divided into two groups: control group)20 head and Fusarium toxins group)30 head in control group and Fusarium toxins group)30 head in Fusarium toxins group)30 group. The control group was fed with basic diet. The Fusarium toxin group was fed with moldy corn and mildew corn protein powder instead of 50% corn and corn protein powder from basal diet. The Fusarium toxin contaminated diet was 0.90 mg/kg Zen 1.43 mg/kg DON 5.85 mg/kg FUMN. The pretest period was 7 days, and the trial period was 35 days. At the end of the trial period, 10 piglets were randomly selected for slaughter sampling. The serum enzyme activity of weaned piglets in Fusarium toxin group was significantly increased, the protein synthesis and antioxidation ability of liver were significantly decreased (P 0.05), the changes of serum antioxidant indexes were consistent with those of liver, and the relative weight of liver was significantly decreased. The expression of inflammatory cytokines IL-1 尾 and IL-6 was significantly increased and the distribution of P0.05 was changed. Pathological observation showed that there were obvious inflammatory cell infiltration and vacuolar degeneration in the liver at the edge of hepatic lobule, compared with the control group. The relative weight of spleen of weaned piglets in Fusarium toxin group did not change significantly (P 0.05), but the antioxidant capacity decreased significantly, which was consistent with the change of corresponding indexes in serum and liver, and the expression of inflammatory cytokines IL-1 尾 and IL-6 was significantly increased and the distribution was changed. Pathological observation showed that the white medullary area of spleen was obviously smaller, the red pulp area was dilated, and vacuolar degeneration occurred locally. In order to study the natural recovery of liver and spleen injury in weaning piglets after stopping feeding with Fusarium toxin contaminated diet, After the first part was slaughtered, 10 piglets were randomly selected from the Fusarium toxin group as natural recovery group. After 21 days, all piglets were slaughtered and sampled. The results showed that the serum enzyme activity of weaned piglets was significantly lower than that of Fusarium toxin group. The protein synthesis ability of liver was improved obviously, but it did not return to the control level, and the antioxidant ability of liver and spleen was improved, which was consistent with the change of serum antioxidant index. The expression of inflammatory cytokines IL-1 尾 and IL-6 in liver and spleen were significantly decreased, and there was no significant difference between the two groups. The above studies showed that the liver function of weaned piglets could be changed by Fusarium toxin 0.90 mg/kg Zen 1.43 mg/kg DON 5.85 mg/kg FUM. The decrease of anti-oxidation ability of liver and spleen and the abnormal expression of inflammatory cytokines in liver and spleen resulted in liver and spleen injury. But it is not completely back to normal.
