不同海拔豬種EPO基因核苷酸多態(tài)性研究

【摘要】:青藏高原低氧脅迫響應(yīng)下,藏豬在長期進(jìn)化過程中形成了獨特的高原低氧適應(yīng)性。本研究對藏豬(合作豬,甘孜藏豬,迪慶豬,西藏藏豬)、八眉豬、煙臺黑豬及長白豬、杜洛克豬、大白豬促紅細(xì)胞生成素(erythropoietin,EPO)基因全序進(jìn)行了測定分析。所得數(shù)據(jù)表明:9個豬群體的37個核苷酸序列共得到14個單倍型,25個變異位點包括8個轉(zhuǎn)換,3個顛換和一個連續(xù)14 bp的缺失,其中有義突變3個;引入豬種僅分布在兩個單倍型H1和H2中,我國地方豬種則分布在13個單倍型中;分析發(fā)現(xiàn)藏豬"TTGGGAAGTCTCAT"的一個序列片段缺失,這一特征在低海拔群體中不存在。EPO基因編碼的194個氨基酸中發(fā)現(xiàn)3個突變位點,變異后的單倍型及氨基酸序列在藏豬中占優(yōu)勢。因此,在適應(yīng)高寒低氧環(huán)境的生理學(xué)和分子學(xué)機制方面,藏豬EPO基因在長期進(jìn)化過程中可能起著關(guān)鍵性遺傳作用。
[Abstract]:In response to hypoxia stress in the Qinghai-Tibet Plateau, Tibetan pigs developed a unique adaptation to high altitude hypoxia during their long-term evolution. In this study, Tibetan pigs (Hehe pig, Ganzi Tibetan pig, Diqing pig, Tibetan Tibetan pig, Bamei pig, Yantai black pig and Landrace pig) were studied. Duroc pigs, The total sequence of erythropoietin erythropoietin (EPO) gene in large white pigs was determined and analyzed. The data showed that there were 14 haplotypes in 37 nucleotide sequences of 9 pig populations, 25 mutation sites including 8 conversions, 3 transversions and a continuous deletion of 14 BP. Among them, there were 3 sense mutants, only two haplotypes H1 and H2 were introduced into pig breeds, and 13 haplotypes were found in native pig breeds in China, and a sequence deletion of "TTGGGAAGTCTCAT" in Tibetan pigs was found by analysis. Three mutation sites were found in 194 amino acids encoded by EPO gene in low altitude population, and the haplotypes and amino acid sequences were dominant in Tibetan pigs. The EPO gene may play a key genetic role in the long-term evolution of Tibetan pigs in the physiological and molecular mechanisms of adaptation to alpine and hypoxic environments.
