【摘要】：泰地羅新(Tildipirosin,TIP)是一種新型的大環(huán)內酯類半合成抗菌藥,是泰樂菌素的衍生物。泰地羅新是一種廣譜、高效的抗微生物藥,對引起豬呼吸道疾病的病原菌,如胸膜肺炎放線桿菌、多殺性巴氏桿菌、支氣管敗血波氏桿菌、副豬嗜血桿菌等,以及引起牛呼吸道疾病的病原菌,如溶血性曼海姆菌、多殺性巴氏桿菌、睡眠嗜組織菌等具有特效,是治療呼吸道疾病極為理想的藥物。目前,在我國還未上市,屬于正在研究中的新獸藥。本研究建立了以泰樂菌素為內標的液質聯(lián)用檢測方法,并完成了泰地羅新注射液在豬體內的藥代動力學研究。檢測方法的質譜條件:采用電噴霧離子源(ESI)正離子模式,檢測方式為多反應監(jiān)測(MRM)掃描模式,噴霧電壓:5.5 k V,離子源溫度(TEM):600℃,氣簾氣(CUR)壓力:20 psi,霧化氣(GS1)壓力:35 psi,輔助加熱氣(GS2)壓力:40 psi,碰撞氣(CAD)壓力:9 psi。泰地羅新和泰樂菌素用于定量的離子分別為:m/z→735.1/98.0和m/z→917.6/174.4。色譜條件:色譜柱為SB-C18(2.1mm×150 mm,3.5μm)色譜柱,以乙腈-0.1%甲酸水溶液(v/v=1:9)為流動相,按0 min(10:90),1 min(10:90),6 min(60:40),6.4 min(95:5),12 min(95:5),12.1 min(10:90),18 min(10:90)進行梯度洗脫,流速0.25 m L/min,柱溫為20℃,進樣量10.0μL。樣品用乙腈提取,氮氣吹干,流動相溶解制備待檢樣品。泰地羅新在0.005-0.8μg/m L的濃度范圍內呈現(xiàn)良好的線性關系,相關系數(shù)為0.9994,檢測限為0.002μg/m L,定量限為0.005μg/m L,該方法的平均回收率在86.96%-95.52%,日內和日間變異系數(shù)均小于15%。且泰地羅新在室溫、-80℃長期冷凍及反復凍融條件下的穩(wěn)定性均良好,符合生物樣品的分析要求。該實驗方法靈敏度高,操作簡便,可以用于泰地羅新在豬體內的藥代動力學研究。藥代動力學結果:通過給豬肌肉注射2、4和8 mg/kg bw三個不同劑量組,用DAS 3.0(Bio Guider Co.,Shanghai,China)軟件非房室模型對數(shù)據(jù)進行分析。其主要的藥代動力學參數(shù)如下:達峰時間Tmax分別為0.208±0.165、0.389±0.18、0.236±0.153 h,峰濃度Cmax分別為0.475±0.114、0.972±0.391、0.987±0.164μg/m L,半衰期t1/2分別為118.69±33.78、124.45±42.06、82.73±23.69 h,藥時曲線下面積AUC分別為11.04±1.27、15.75±5.19、37.54±4.83μg/m L*h。以上實驗數(shù)據(jù)為泰地羅新注射液在臨床合理用藥提供了理論依據(jù)。
[Abstract]:TildipirosinTIP) is a new type of macrolide semi-synthetic antimicrobial agent, which is a derivative of tylosin. It is a broad-spectrum, highly effective antimicrobial agent against pathogens causing respiratory diseases in pigs, such as Actinobacillus pleuropneumoniae. Pasteurella multocida, Pasteurella bronchus, Haemophilus parais, and other pathogens that cause bovine respiratory diseases, such as Mannheim haemolyticus, Pasteurella multocida, histiophilic sleep bacteria, etc., have special effects.
