2014年-2016年廣西地區(qū)豬繁殖與呼吸綜合征病毒Nsp2和ORF5基因的遺傳多樣性分析

【摘要】:為了解廣西地區(qū)豬繁殖與呼吸綜合征病毒(PRRSV)流行毒株的遺傳進(jìn)化情況,對(duì)2014年-2016年來(lái)自廣西各地的部分PRRSV陽(yáng)性病料進(jìn)行Nsp2和ORF5基因的擴(kuò)增和測(cè)序分析。結(jié)果獲得34個(gè)Nsp2基因序列和45個(gè)ORF5基因序列,均屬于美洲型毒株。Nsp2基因間核苷酸序列的同源性為91.8%~100%,與PRRSV美洲型毒株VR-2332、CH-1a、JXA1及NADC30株核苷酸序列的同源性分別為81.3%~84.3%、88.9%~92.1%、94.3%~99.3%和73.5%~75.1%,而與PRRSV歐洲型毒株LV株核苷酸序列的同源性為51.5%~53.2%。ORF5基因間核苷酸序列的同源性為82.8%~100%,與PRRSV美洲型毒株VR-2332、CH-1a、JXA1及NADC30株核苷酸序列的同源性分別為83.7%~99.5%、85%~95%、83.8%~99.7%和83.2%~86.4%,而與PRRSV歐洲型毒株LV株核苷酸序列的同源性為62.4%~64.5%;贜sp2和ORF5基因推導(dǎo)的氨基酸序列繪制的遺傳進(jìn)化樹中,廣西地區(qū)的毒株主要分布在以JXA1為代表的Ⅳ亞群。表明當(dāng)前廣西PRRSV流行毒株以JXA1株為代表的高致病性美洲型毒株為主,各毒株Nsp2和ORF5基因序列存在一定的差異,尚未發(fā)現(xiàn)歐洲型毒株和美洲型NADC30類毒株。
[Abstract]:To understand the genetic evolution of porcine reproductive and respiratory syndrome virus (PRRSVV) epidemic strains in Guangxi. From 2014 to 2016, some samples of PRRSV positive venereal diseases from various parts of Guangxi were amplified and sequenced by Nsp2 and ORF5. Results 34 Nsp2 gene sequences and 45% Nsp2 gene sequences were obtained. A sequence of ORF5 genes. The homology of nucleotide sequence between the two strains was 91.8100, which was similar to that of PRRSV strain VR-2332 (CH-1a). The nucleotide sequence of JXA1 and NADC30 strains were 81.3% and 84.3%, respectively. 94.3% and 73.5% respectively. The nucleotide sequence homology with LV strain of PRRSV was 51.5%. The homology of ORF5 gene sequence was 82.8%. The homology of nucleotide sequence with PRRSV strain VR-2332CH-1aA1and NADC30 strain was 83.7% and 99.585%, respectively. 83.8% and 83.2%, respectively. The nucleotide sequence homology with PRRSV European strain LV strain was 62.4% and 64.5%. The amino acid sequences derived from Nsp2 and ORF5 genes were mapped in the phylogenetic tree.
